Crowdfunding a jeho potenciál v EU a ČR

This diploma thesis deals with the development and potential of crowdfunding in the European Union and the Czech Republic. The thesis examines the structure of campaigns published on two crowdfunding portals - Kickstarter and HitHit. The main part is making of a logit model, which result is evaluation of the factors that influence the success of the campaigns. The examined factors include the number of supporters, the category to which the campaign is assigned, or the target amount. The main aim of this thesis is evaluation of the potential of crowdfunding in the Czech Republic and the European Union. The thesis also deals with evaluati-on of successful crowdfunding campaigns. At the end of the thesis there are evaluated success factors, such as a dance category or projects originating by initiators from the Great Britain. There are also discussed some risks which are associated with this type of financing and may threaten future development of crowdfunding in the European Union, such as Brexit.
Characteristics of expression vectors for Corynebacterium glutamicum and their use for studies of sigma factors of RNA polymerase

The aim of the thesis was to characterize chosen expression vectors used in biotechnologically important bacterial species, Corynebacterium glutamicum, and to test their use in studies of promoter activity control by sigma factors of RNA polymerase. Different properties of these vectors (level of expression of the cloned gene, leaky expression without inducer, dependence of expression level on inducer concentration and cell population homogeneity) were found by determination of expression level of the model gfpuv gene by fluorescence intensity assay of the produced protein and by gfpuv-expressing C. glutamicum cell population analysis using flow cytometry. The vector pEC-XT99A was chosen for testing the bi-plasmid system for assignment of a sigma factor to the chosen promoter. Although the level of expression provided by pEC-XT99A was not high, the vector showed no leaky expression, expression from the vector was comparable for a wide range of IPTG concentrations and the cell population was homogenous concerning the gene expression. Title: Analysis of the game performance in relation to different types of the match mini-basketball players. Objectives: The objective of this study is analysis and comparison of the selected performance indicators of the player in relation to the current competiton system in the Czech minibasketball (5x5 vs 3x3 with one basket). Methods: As a research method, the method of observation of the video recordings. Using the basic statistic and arithmetic methods, the measured data were processed statistically and consequently, graphically also. Results: It was discovered that frequency of selected performance indicators is higher , for the whole team as well as individual players in form of modified minibasketball 3x3, in most cases. Functions of sigma factors of RNA polymerase in Corynebacterium glutamicum

The aim of this thesis was to characterize the function of sigma factors of the bacterium Corynebacterium glutamicum and to analyze the promoter sequences which are recognized by individual sigma factors. Sigma (σ) factors are the subunits of RNA polymerase, which allow recognizing the sequences of specific promoter regions of the gene and initiating its transcription. The C. glutamicum genome carries genes encoding the primary sigma factor σA and six alternative sigma factors, σB , σC , σD , σE , σH and σM , whose expression is changed depending on the growth conditions and in response to the stimuli from the surrounding environment. Regulation of gene expression at the level of transcription is one of the mechanisms of the adaptation of cells to changes of living conditions. At the conclusion of this work, a model of the regulatory network of sigma factors, which is a core of the complex regulatory network controlling all processes in the cell, is proposed. Child labour in developing countries Abstract This bachelor thesis deals with the child labour and its occurence in developing countries. The main aim is to present the basic view of this problem. The term of child labour relies here on Convention on the Rights of the Child and conventions of International Labour Organization. There are several types of child labour, in which children appear most, including the worst forms of child labour. Every type includes description of activities performed and their consequences on children. Child labour is influenced by society and family, demand for child labour, economics shocks and the lack of access to education. All mentioned causes of child labour are related to poverty. Selected solutions of child labour are divided into assistance from the state, employer, international organizations, govermental projects and nongovermental organizations. Nursing care of patients with lues in the isolation mode of Dermatology and Venerology Department.

The bachelor thesis "Nursing care for patients with lues in the isolation mode of dermatovenerological department" is divided into the theoretical and research part. The theoretical part deals with lues diseases , it´s examination methods which are used to prove this desease. It is also focused on the treatment of patients with lues - divided into complex and medical treatment. Part of the treatment is also dispensary care and duty to report of sexual illness after hospitalization of patients which are performed in the outpatient department of dermatovenerology department. Another part of the theoretical part is focused on nursing care in lues patients in isolation and satisfying their needs. The communication with the patients is also important. We have to choose the right method of communication to be successful. The research part of this thesis includes a qualitative research performed by non-standardized interview. The first variation of interviews was addressed to general registered nurses who are working on the bed part of the dermatovenerology department. This group consisted of six women. Interviews with general nurses was focused on the nursing care in isolation, satisfying patients needs, communication with them and dealing with the problem situations which may occur during taking care of patients in isolation. The second group consisted of patients who were hospitalized in the isolation (three men and three women). Interviews with this group of people were focused on their knowledgeableness of their illness, treatment, the isolation, how they perceive the provided care, the level of satisfying their needs and the perception of communication with the nursing staff. The aim of the work was to find out the specifics of nursing care for patients with lues in isolation from the perspective of nurses. The secondary aim was to find out how patients with lues perceive providing nursing care. The research has shown that the care for the patients in isolation is less physically demanding because the patients are more likely young adn self-sufficient. Much more difficult - according to the nurses - is observe the strict rules of barrier care which is necessary in isolation. Specific activities which are provided during nursing are not perceived as burden. On the other hand the nurses perceive the communication with the patients as difficult because some of them are maladjusted and sometimes they are verbally aggressive. The nurse has to find different communication method for each patient. The second part of the research dealt with the patient´s perception of nursing care. Research results from my survey shown that patients are satisfied with the nursing care (including the safisfying their needs). Only minor deficiencies were uncovered by two patients. They imagined that the provided nursing care would be more extensive. Patients reported that they perceived their illness negatively and with fear. Finally, I can state that the provided care is part of healing, islolation regime which is necessary for hospitalized patients. In spite of minor problems patients are dealing well with the isolation. Compared with the standard room the specifics of nursing care in isolation is not find to be burden.

It also deals with transkarbam 12 and its mechanism of action, transdermal application of active substances and electrical resistance as a physical unit. Experimental part of this work describes setup used for permeation in vitro experiments on pig ear skin of full thickness. The aim of this work was to find experimental evidence and interpretation of relation between electrical resistance of the skin and flux of caffeine from donor suspension or creams containing transkarbam 12 and also evaluate feasibility of the correction of the variability of the permeation experiments results using preceding infinite dose experiment. In theoretical part of this work, the skin barrier function and transdermal permeation enhancers (with focus on Transkarbam 12, T12) were reviewed. In following experimental part, the acceleration effect of various concentrations of Transkarbam 12 (T12) on in vitro caffeine permeation through full-thickness pig ear skin was evaluated. Caffeine was administered in oleocreams containing 30 % aqueous phase. The values of mean flux of caffeine Jr from creams containing 0%; 0,3%; 0,5%; 0,7% and 1% T12 were 6,29 ± 0,90 μg.cm-2.h-1; 9,13 ± 0,95 μg.cm-2.h-1; 8,71 ± 1,91 μg.cm 2.h-1; 10,20 ± 3,25 μg.cm-2.h-1 and 7,06 ± 0,81 μg.cm-2.h-1 respectively. The optimization of system of employees´ benefits for non-profit organization according to the needs of employees and possibilities of employer

The diploma thesis is focused on the optimization of the system of employees´ benefits in the foundation Dobrý Anděl. The first part of the thesis is aimed at a segmentation of employees´ benefits from different aspects, especially tax deductibility and non-deductibility of expenses for the employer and the payment liabilities to the employee. The second part of the thesis deals with the concrete proposal of the new system of benefits. This proposal is created based on the financial possibilities of employer and also on the expressed preferences in individual benefits of employees. Preferences of employees are determined on the basis of written survey. From these results is created a realistic proposal of the system of benefits for foundation Dobrý Anděl, which also consider the possibility of future expansion of the whole organization. The newly created system of benefits is financially viable for the foundation and also counts with the financial reserve, which is necessary for the non-profit organization, because all the funds for operating expenses are obtained from donations.
