Metodika je zaměřena na konzervování-restaurování nízkopálené nestabilní keramiky. Podává ucelený přehled o vhodných postupech pro zpracování materiálu tohoto typu. Obsahuje souhrn vhodných metod pro hodnocení stupně poškození, postupy odběru vzorku a doporučené analýzy pro identifikaci chemického a mineralogického složení keramiky. Dále popisuje možnosti čištění, postupy konsolidace, lepení a doplňování chybějících fragmentů a případné retuše. Obsahuje rovněž zásady pro vhodné uložení těchto předmětů a pro pochopení jednotlivých metod jsou uvedeny komentované příklady.

Metodika je zaměřena na konzervování-restaurování glazované keramiky s ohledem na stabilitu a pórovitost keramického střepu. Podává ucelený přehled o vhodných postupech pro zpracování materiálu tohoto typu. Obsahuje přehled vhodných metod pro hodnocení stupně poškození (jak střepu, tak glazury), postupy odběru vzorku a metody pro identifikaci chemického a mineralogického složení keramiky i glazury a stability jejich celkového systému. Dále popisuje možnosti čištění, postupy konsolidace, lepení a doplňování chybějících fragmentů a případné retuše střepu i glazury. Obsahuje rovněž zásady pro vhodné uložení těchto předmětů a pro pochopení jednotlivých výsledků jsou uvedeny komentované příklady.

Application of the Property Right to Real Estates Transfer Abstract The aim of this thesis is to analyze the provisions relating to the transfer of ownership of real estate, together with practical examples referring to the diverse case law and other decisions that were created not only by the judicial system. Apart from the provisions of the Civil Code, I also focused on the provisions of other legal regulations, especially on the provisions of the Cadastr Act, which is a crucial regulation for the transfer of ownership. In addition to other national regulations, I also pointed out the importance of regulation of property rights in international legal context and focused on the foreign legal regulation of specific steps in the transfer of real estate. Despite the differences in the legal regulations of foreign countries, the legal regulation of the procedure for the transfer of real estate was very similar, following our membership within the European Union, which guarantees us the same rights as the citizens of those countries. It also deals with transkarbam 12 and its mechanism of action, transdermal application of active substances and electrical resistance as a physical unit. Experimental part of this work describes setup used for permeation in vitro experiments on pig ear skin of full thickness. The aim of this work was to find experimental evidence and interpretation of relation between electrical resistance of the skin and flux of caffeine from donor suspension or creams containing transkarbam 12 and also evaluate feasibility of the correction of the variability of the permeation experiments results using preceding infinite dose experiment. The relationship between lessee and lessor compared to Airbnb service Abstract The aim of this diploma thesis is to analyze the legal aspects of lease pursuant to the general and special provisions of the Civil Code as well as to point out the different and currently ambiguous legislation of the Airbnb online service. As the lease provisions are strongly protectionist in relation to the lessee, who stands as a weaker party to the lessor, I also elaborate on the fundamental rights and obligations of each of the parties. Not only the Civil Code together with other national provisions, which now regulate in detail most institutes related to lease, represent a basic source of the diploma thesis, but also an unforgettable foreign legislation, which is especially with regard to the Airbnb service a very interesting source of inspiration for the future development not only in the Czech Republic. The content itself is divided into five parts. The first of them defines the lease according to the general provisions of the Civil Code together with its essential characteristics, which thus enables an easier understanding of the special regulation of the residential lease. Crowdfunding a jeho potenciál v EU a ČR

This diploma thesis deals with the development and potential of crowdfunding in the European Union and the Czech Republic. The thesis examines the structure of campaigns published on two crowdfunding portals - Kickstarter and HitHit. The main part is making of a logit model, which result is evaluation of the factors that influence the success of the campaigns. The examined factors include the number of supporters, the category to which the campaign is assigned, or the target amount. The main aim of this thesis is evaluation of the potential of crowdfunding in the Czech Republic and the European Union. The thesis also deals with evaluati-on of successful crowdfunding campaigns. At the end of the thesis there are evaluated success factors, such as a dance category or projects originating by initiators from the Great Britain. There are also discussed some risks which are associated with this type of financing and may threaten future development of crowdfunding in the European Union, such as Brexit.
Characteristics of expression vectors for Corynebacterium glutamicum and their use for studies of sigma factors of RNA polymerase

The aim of the thesis was to characterize chosen expression vectors used in biotechnologically important bacterial species, Corynebacterium glutamicum, and to test their use in studies of promoter activity control by sigma factors of RNA polymerase. Different properties of these vectors (level of expression of the cloned gene, leaky expression without inducer, dependence of expression level on inducer concentration and cell population homogeneity) were found by determination of expression level of the model gfpuv gene by fluorescence intensity assay of the produced protein and by gfpuv-expressing C. glutamicum cell population analysis using flow cytometry. The vector pEC-XT99A was chosen for testing the bi-plasmid system for assignment of a sigma factor to the chosen promoter. Although the level of expression provided by pEC-XT99A was not high, the vector showed no leaky expression, expression from the vector was comparable for a wide range of IPTG concentrations and the cell population was homogenous concerning the gene expression. Using pEC-XT99A from which individual stress sig genes were expressed, the σD factor was clearly assigned to the up-to-now unknown Pcg0420 promoter. Title: Analysis of the game performance in relation to different types of the match mini-basketball players. Objectives: The objective of this study is analysis and comparison of the selected performance indicators of the player in relation to the current competiton system in the Czech minibasketball (5x5 vs 3x3 with one basket). Methods: As a research method, the method of observation of the video recordings. Using the basic statistic and arithmetic methods, the measured data were processed statistically and consequently, graphically also. Results: It was discovered that frequency of selected performance indicators is higher , for the whole team as well as individual players in form of modified minibasketball 3x3, in most cases. Functions of sigma factors of RNA polymerase in Corynebacterium glutamicum

The aim of this thesis was to characterize the function of sigma factors of the bacterium Corynebacterium glutamicum and to analyze the promoter sequences which are recognized by individual sigma factors. Sigma (σ) factors are the subunits of RNA polymerase, which allow recognizing the sequences of specific promoter regions of the gene and initiating its transcription. The C. glutamicum genome carries genes encoding the primary sigma factor σA and six alternative sigma factors, σB , σC , σD , σE , σH and σM , whose expression is changed depending on the growth conditions and in response to the stimuli from the surrounding environment. Regulation of gene expression at the level of transcription is one of the mechanisms of the adaptation of cells to changes of living conditions. At the conclusion of this work, a model of the regulatory network of sigma factors, which is a core of the complex regulatory network controlling all processes in the cell, is proposed.
