Prof. PharmDr. Jaroslav Roh, Ph.D. Title of Doctoral Thesis: Synthesis of purine derivatives with potential antimycobacterial activity The theoretical part of this dissertation briefly summarizes basic data on tuberculosis (TB), an infectious disease caused by Mycobacterium tuberculosis (Mtb.). The epidemiological data published in the annual report of the World Health Organization (WHO) and the structure of drugs used in current TB therapy are presented. The theoretical section also gives a brief overview of the treatment of TB and mentions four compounds that are currently in phase II clinical trials. The mechanism of action of these compounds is inhibition of the enzyme decaprenylphosphoryl-β-D-ribose-2'-epimerase, specifically the DprE1 subunit. This enzyme plays an important role in the synthesis of mycobacterial call wall. The next part of the dissertation summarizes the results of research on potential purine and pyrrolo[3,2- d]pyrimidine antituberculosis agents derived from purine-6-one 10. Based on the theoretical part and the analysis of the current situation are proposed solutions suitable for the management of access network provider. The proposal part contains selection of the best proposal and its implementation into the routine mode of operation.
Production of ethanol as an automotive fuel from renewable materials

The thesis deals with ethanol production using renewable energy ressources. It describes various manufacturing processes and the usage of respective input materials. It researches utilization of ethanol as engine fuel both as a mixture with conventional fuels and also as the principal component. Consequentionally, it proposes a design of combustion engines suitable for this fuel. Evaluation of advantages and disadvantages of substituting petrol products with bioethanol is provided. The thesis also includes energy and economic evaluation.
Sewage disposal plant

This thesis deals with the design and static assessment of a concrete part of a sewage treatment plant. Specifically, the calculation and assessment of the base plate, external walls, internal walls and ceiling slabs, elaboration of shape and reinforcement drawings of selected structural element. The concrete part of the Sewage disposal plant is all below ground level. Static system and the calculation of internal forces was carried out in Scia Engineer 2011 - student version.
Commented translation of a text on science and technology

Tato bakalářská práce se zabývá používáním technického jazyka v češtině i angličtině, v čem si jsou tyto jazyky podobné a v čem se naopak liší. Pro studování jazyků v elektrotechnice byl vybrán technicky zaměřený text k přeložení a poté byl přeložen do angličtiny. Po překladu byla provedena analýza jazyka, které se zaměřovala jak na lingvistické, tak extralingvistické vlastnosti, které jsou často specifické pro odborné texty. Tyto vlastnosti pokrývají širokou škálu lingvistických oborů, jako jsou například lexikologie, morfologie, stylistika, syntax, ale i extralingvistické záležitosti, jako třeba pragmatika. Analýza originálního a přeloženého textu odhalila spoustu rozdílů, jak malých, tak i velkých, které jsou výsledkem pokusu o přenos informace v jazyce, který je poměrně rozdílný oproti originálnímu jazyku.

The thesis is to analyse and design universal cabling in a multi-functional house that will be subject to a thorough reconstruction. The house shall serve for business purposes of the company GNT, s.r.o.. The cabling design is based on investor´s requirements and the construction plan analysis of the house. There had been no computer network in the house. The thesis shall encompass a complete documentation of the project, and its estimated budget.
The road bridge

This master' thesis deal with the desiggning of steel bearing structure of road bridge with span of 48,5m with lower bridge deck, which cross local road over the river Thaya in the town Znojmo. Three variants have been designed of arched bridge. The most economical variant was prepared in detail and wascarried out static assessment of individual elements and selected details. In detail solved variant is the bridge with free staing arches and lower bredge deck with composite crosspiece. The span of bridge is 48,5m, width of bridge is 11,5meters, free width of main trafic area 6,5m. Camber of bridge 8,0m
Methodical Approach to Evaluate the Efficiency of Road Infrastructure Construction using the PPP Projects

This diploma thesis analyses and elaborates a methodical tool for evaluating the advantage of realization of construction of road infrastructure in the form of a PPP project. Furthermore this tool is subsequent applied to a particular project. The introductory part summarizes the basic theoretical knowledge regarding the project financing in the form of public-private partnerships. In the following part, a methodological tool for comparing the value for money based on different methods of realization. In the last part of the diploma thesis, this methodical tool is applied to a specific project, including a basic feasibility study.
Study of alkali metals ions diffusion in hardened cement

This master thesis is aim to study alkali diffusion (sodium, potassium) in hardened cement paste body. It´s deal with impact of various sorroudings (hydroxide, nitrate and carbonate) in various concentrations and temperature effect. This work is also deal with changes in cement paste and medium. Teoretic part is talking about cement production and cement chemistry, alkali-silica reaction, plasticizer, diffusion an cement testing metods. This work is aimed to white (Aalborg CEM I 52,5 R – SR 5) and road (Mokrá CEM I 42,5 R – sc) cement. Experimental part describes constitution of tested cement pastes, conservation of them and sampling. Diffusion is study by ICP-AES which affected the loss of alkali from the solution and SEM which affected alkali migration (diffusion) in cement paste. In several cases, the formation of a saturated alkali edge was observed, followed by steady diffusion. In the case of potassium there was very slow diffusion through the body mass. The influence of medium and temperature was evaluated as the most significant. The effect of medium concentration wasn´t significant. Also changes in the porosity on the body were observed. The diffusion itself was affected by secondary products gorwing on the surface of body and in the medium. The work deals with them in the last part.
DNS firewall and its deployment and integration in cyber center

This bachelor's thesis deals with the deployment, integration, and testing of a DNS firewall in a security operations center. It describes the connection of endpoints and remote local area networks to the DNS firewall located in the security operations center. Furthermore, the enforcement of the DNS firewall is described. The main goal of the thesis was to deploy and integrate a DNS firewall inside a security operations center. The first chapter describes the security operations center in general. The second chapter deals with the DNS system. The following chapter describes the security of the DNS system and security of DNS requests, the reader is informed of the term DNS firewall and RPZ and VPN technologies. The fourth chapter describes the DNS firewall deployment process and its integration in a real security operations center. The next chapter describes connection methods of endpoint and remote local area networks to the DNS firewall and its enforcement inside the security operations center. The last chapter deals with performance testing and deployed DNS firewall availability. The outcome of the thesis involves a deployed, integrated, fully-functional, and tested DNS firewall in a real-world security operations center. The Bind software package along with the RPZ technology was used to implement and deploy the DNS firewall. For testing and connection of endpoints, the VPN technology, and the RIPE Atlas network was used.
