Midwife Care for Woman with Endometriosis

This bachelor thesis deals with the topic of midwife care for a woman with endometriosis. The thesis is divided into theoretical and practical part. The theoretical part is mainly focused on the endometriosis disease. The theoretical part describes the definition of endometriosis, its causes, diagnosis, manifestations, treatment options and midwife care. For the practical part, two objectives were chosen. The first aim was to find out how women with endometriosis perceive midwife care. The second aim of the thesis was in turn to find out how midwives perceive the whole issue of this disease. The bachelor thesis had three research questions. The first research question focused on how women with endometriosis perceive midwives' care. The second research question explored how the lifestyle of women with the condition has changed. The third research question looked at how midwives perceived the issue of endometriosis. The aforementioned objectives were achieved through a qualitative research investigation in the form of semi-structured anonymous interviews. Data collection for the research inquiry took place from January to March 2023. In the practical part, two research sets were chosen. The first research set consisted of six midwives who encountered women who had been diagnosed with endometriosis during their practice. To preserve their anonymity, the midwives were referred to as PA1 to PA6. The second research group consisted of eight women with endometriosis who were referred to as Ž1 to Ž8 in the thesis. The data collected was transcribed and then further analyzed using text coloring. Main categories and sub-categories were established for data analysis, these are further described in the research section of the thesis. The first research set consists of two main categories and four subcategories. Two main categories and six subcategories were obtained from the informants' statements of the second research set.
A fairy tale as means to preschool child development

The bachelor thesis is focused on working with an authored fairy tale with preschool children in a kindergarten environment. The theoretical part contains three basic chapters. The first chapter introduces the characteristics, historical development and basic classification of fairy tales. It summarizes the function of fairy tales in preschool education and discusses the language style of fairy tales. The second chapter is focused on the fairy tale as a tool in preschool education. The third chapter deals with the analysis of the Preschool education curriculum framework (PECF) and the learning domains. The practical part deals with the implementation of philosophical discussion, creative writing and artistic elaboration of the authored fairy tale. Furthermore, it deals with the research investigation of the influence of a fairy tale story on a group of children in kindergarten.
The use of information technologies in preschool children from the perspective of kindergarten teachers

The final work entitled "The use of information technology in preschool children from the perspective of kindergarten teachers" deals with information technologies used by preschool children, how they work with them and how their use is reflected in preschool education. The theoretical part of the bachelor thesis contains chapters on the development and history of information and communication technologies, the impact of information technology on children and youth, the risks and benefits of using information technology. In the practical part, based on mixed research (observation, questionnaire survey and interviews with kindergarten teachers), research questions related to the goal of the bachelor's thesis are answered.
Nanoparticle-Mediated Delivery System for Gene Therapy

Gene editing with the CRISPR/Cas9 system is one of the options that sets a new trend in the development of gene therapy. The most commonly used delivery of DNA into the cells are via viruses. Nevertheless, they are often unable to take CRISPR/Cas9 system, which can be bigger than several kb. Nanoparticles (NPs), as non-viral transporters, seem to be a good alternative delivery system. For this work magnetic Fe3O4 NPs (MNPs) were selected, because of their excellent properties such as multifunctionality, biocompatibility, easy degradation and simple synthesis. The aim of this work was to synthesise MNPs and a complex of MNPs coated with PEI/CRISPR-Cas9 plasmid and to characterize them by physicochemical methods. The created complex MNPs/PEI/CRISPR-Cas9 was defined by exact parameters that are suitable for possible cell uptake. The hypothesis of stabilization of the MNPs/CRISPR-Cas9 plasmid complex by polyethylenimine (PEI), which can also protect plasmid DNA against restriction endonucleases, was verified. Next a stable modified cell line HEK293-TLR3, designed to evaluate the efficacy of double strand break (DSB) repair by nonhomologous end joining (NHEJ) or homologous recombination (HR) was, transfected with the synthesised MNPs/PEI/CRISPR-Cas9 complex. Christianization of early medieval society in the view of the movable archeological sources

The presented text deals with a process of Christianization with taking account of informative value of archaeology. Movable archaeological artifacts generally connected with Christianity are paid particular attention. These objects were divided into two groups: Group I - artifacts primarily linked to Christianity, Group II - artifacts with practical or decorative function carrying Christian theme. One category of objects was chosen from both the groups (Group I - hanging crosses, Group II - Strap end) and analysed. At the end a contribution towards knowledge of the process of Christianization is evaluated.

Benefits of Neoadjuvant Therapy in Colorectal Cancer Treatment

This Bachelor's thesis focuses on benefits of neoadjuvant therapy for colorectal cancer. The thesis is divided into a theoretical and a practical part. The theoretical part contains data I have compiled from available literature, scholarly articles and professional websites. I concentrated most closely on colon and rectal anatomy, screening, diagnosis and treatment of colorectal cancer. The aim of the Bachelor's thesis is to compare the extent of colorectal cancer before and after neoadjuvant therapy. Furthermore, the second aim is to determine the number of radically operated patients after preoperative treatment. The practical part consists of data from the archive of medical records at the Department of Oncology in the České Budějovice Hospital, a. s. The observed factors were age and gender of the patient, diagnosis, localization and TNM classification, type of neoadjuvant therapy, initiation and termination of therapy, type of operation and its initiation. The last factor was the extent of disease after surgery. In total, I evaluated 50 patients who met the criteria. I processed the data into comprehensive graphs and spreadsheets. The collected data show an improvement in the extent of disease in 84 % of patients. 5 % of patients had the same TNM classification and the disease worsened in 11 % of patients. Complete or nearly complete remission occurred in 6 patients (14 %). 88 % of patients from the whole research group were radically operated. 20 patients (40 %) underwent rectal resection and 24 patients (48 %) underwent rectal amputation. 6 remaining patients did not have any surgery. Two patients had an inoperable tumour, others were not operated due to their high age, weight or because it was their wish. The results from the practical part can provide feedback to the Department of Oncology in the České Budějovice Hospital. Next, it can also serve as a study material for students and the general public.
Potential of Prague theatres in domestic and incoming tourism

The main goal of this thesis is to evaluate the fulfillment of the potential of Prague theatre institutions in the domestic and incoming tourism of the Czech republic, to find the most significant Prague theatre institutions and compare the way they present their offer with institutions in competitive destinations. The first two chapters introduce the cultural tourism in general, in the Czech republic and in Prague. The following chapter describes the offer of Prague theatres and the fulfillment of its potential, where the evaluation is based on the results of a research, carried out by the author herself. The last chapter presents a comparison of presentation on websites of chosen theatres in Berlin, Dresden and Vienna with Prague.
Generation of large animal models using genome editing

The principle of gene engineering is the intervention to the DNA of the studied organism. After the discovery of the programmed endonucleases, there has been a great expansion of this technique and it also accelerated the possibilities to create large animal models. Until recently, large animal models were very difficult to be generated. These endonucleases include zinc finger nuclease (ZFN), transcription activator like effector nuclease (TALEN) and CRISPR/Cas9. All endonucleases produce locally specific splicing in the targeted segment of the genome. This splicing is most easily corrected by the non-homologous ends joining (NHEJ), so then it is possible to create a so -called knock-out (KO) model. The second type of repair is homologous recombination (HR) using a DNA template with homologous arms. This makes it possible to create a knock-in (KI) model that cannot be created without specific endonucleases in large animal models due to the low natural HR. This work summarizes the history, technique and the use of programmed endonucleases for the creation of large animal models. These models have a great use in biomedicine, mostly in preclinical research, they are also significant in agriculture and even in the environment protection.
