The optimization of SPME for the determination of aroma compounds in sugar cane rums

This master’s thesis deals the optimization of SPME for the determination of aroma compounds in sugar cane rums. The gas chromatograph with mass detector was used for the measurement. The optimization was performed on different type of SPME fibers. This method was used for the identification and the quantification of compounds in selected samples of sugar cane rums purchased from the commercial network.

The bachelor thesis is devoted to important substances in caraway oil. The determination of substances was performed in the laboratories of the Brno Institute of Brewing and Malting, a.s. It is divided into theoretical and experimental part with results and their discussion. The theoretical part describes caraway (Carum carvi), its use and composition of caraway oil, including the possibility of its use. The practical part deals with the identification and determination of content substances in caraway essential oils by the method of gas chromatography and mass spectrophotometry (GC-MS). It also includes a comparison of the content of different caraway essential oils.
Monitoring of gluten content in barley varieties recommended for the production of Czech Beer

Sandwich enzyme-linked immunosorbent assay ELISA was used for determination of gluten in barley varieties recommended for the production of Czech beer. Currently, it is the most widely used method for determining gluten in the food industry. Monitoring the gluten content in raw materials and foods made from them is especially important for people suffering from celiac disease. Gluten intolerance is an autoimmune disorder that causes morphological and functional changes in the small intestine. Currently, the most effective treatment is following a gluten-free diet. In this bachelor thesis, content of gluten in 14 barley samples was monitored. The RIDASCREEN® Gliadin set was used for the measurement. Results were evaluated with RIDA SOFT Win.NET software. Content of gluten in the barley samples varied from 36,3 g/kg to 82,5 g/kg.

Fenolické látky vo vybraných rastlinných druhoch

This diploma thesis deals with the analysis of phenolic compounds such as phenolic acids and flavonoids in selected plants, such as ribwort plantain (Plantago lanceolata L.) in which the content of chlorogenic acid, caffeic acid, verbascoside, ferulic acid, rutin and quercetin was monitored. The content of chlorogenic acid, coffee acid, routine, hyperoside, isoquercitroside, tiliroside, quercetin and kaempferol was monitored in small-leaved lime (Tilia cordata Mill.) In addition, the content of chlorogenic acid, caffeic acid, coumaric acid, rutin, hyperoside, isoquercitroside, quercetin and kaempferol was monitored in marigold (Calendula officinalis L.). Also the content of chlorogenic acid, coffee acid, rutin, hyperoside, isoquercitroside, quercetin and kaempferol was monitored in elderberry (Sambucus nigra L.). Similarly the extraction of phenolic compounds from these plants was carried out and subsequently subjected to chromatographic analysis on a UPLC liquid chromatograph with a PDA detector at the Research Institute of Brewing and Malting, Plc.

Bachelor theses is themed "Variability in the content of essential oils in selected plants used in herbal medicine, aromatic plants and spice plants" and deals with the variability of the content of essential oils in caraway (Carum carvi L.), where carvone and limonene are present, in chamomile (Matricaria recutita L.), which contains farnesene, chamazulene, bisabolol oxide A and apigenin-7-glucoside, and in lavender (Lavandula angustifolia), which contains camphor, borneol, alpha-terpineol and linalylacetate. The thesis consists of two parts -- the first one is a literature review of selected plants and the second one is a practical part held in the laboratory at Department of Crop Science, Breeding and Plant Medicine at Faculty of Agriculture. In the practical part, drugs were individually analyzed using the methods taken from Czech pharmacopoeia. Samples from caraway were acquired from Agritec, s.r.o. company's breeding, lavender and chamomile came from the company Leros, s.r.o. and also from domestic production. Variation analysis of caraway proves that content of both examined substances -- carvone and limolene -- was highly affected by its genotype. Total content of essential oils, on the other hand, was not statistically affected by genotype. Four different variants were studied in chamomile. The content of essential oils in chamomile was statistically highly affected by monitored variants; the content of chamazulene was affected in the same way. Bisabolol oxide A was significantly affected by the variant of chamomile and so was apigenin-7-glucoside. Farnesene was not significantly affected by variant. All compounds found in lavender's essential oils, namely camphor, borneol, alpha-terpineol and linalyl-acetate, were statistically greatly affected by variant.
