Anti-cancer drugs with immunoprotective/immunomodulatory effect
Říhová, Blanka ; Etrych, Tomáš ; Kovář, Marek ; Chytil, Petr ; Hovorka, Ondřej ; Plocová, Daniela ; Mrkvan, Tomáš ; Strohalm, Jiří ; Šírová, Milada ; Šubr, Vladimír ; Kubáčková, K. ; Ulbrich, Karel
Cytotoxic drugs based on HPMA polymer show direct cytotoxic and cytostatic effect together with an ability to mobilize the immune system. Two types of linkage of the drug to the polymer are discussed, as well as linkage-related differences in tumour cell accumulation and fate of the cells
Intracellular cleavage of doxorubicin is not a necessary prerequisite for the pharmacological activity of HPMA-based conjugates. Composition of the spacer bearing drug, and the way of doxorubicin binding strongly influences intracellular distribution of polymeric drugs and in that way the mechanism of cell death
Kopolymery na bázi HPMA s pH-řízeným uvolňováním doxorubicinu v protinádorové terapii umožňují imunitnímu systému rozpoznat nádor

Therapy with HPMA-based conjugates triggers effective anti-tumour immunity early during the course of the treatment. Activated immune system, in addition to direct cytotoxic effect the drug, is responsible for the eradication of established tumour
Protinádorová účinnost konjugátu na bázi N-(2-hydroxypropyl)metakrylamidu, obsahujícího doxorubicin a normální lidský imunoglobulin
Šírová, Milada ; Strohalm, Jiří ; Rossmann, Pavel ; Mrkvan, Tomáš ; Šubr, Vladimír ; Plocová, Daniela ; Ulbrich, Karel ; Říhová, Blanka
Polymeric conjugate based on HPMA containing cytotoxic drug (doxorubicin) and human “non-specific” immunoglobulin has a significant anti-tumour effect and does not impair immune functions of the host. C57BL/6 mice with EL-4 lymphoma, when completely cured with the conjugate, were protected against the same tumour due to a specific anti-tumour immune reaction
Memory of the immune system
Říhová, Blanka
Defense against microbes is mediated by the early reactions of innate immunity and the later responses of adaptive immunity.
