Antimicrobial efficacy of quaternary ammonium salts

This dissertation thesis deals with the antimicrobial susceptibility testing of newly prepared compounds based on quaternary ammonium salts (KAS) and their reference standards commonly used in practice. The new compounds were prepared in cooperation with the Department of Toxicology and Military Pharmacy of the Faculty of Military Health Sciences of the University of Defence and the Biomedical Research Centre at the University Hospital Hradec Králové as a part of joint projects. The antimicrobial activity was established on gram-positive (G+) and gram-negative (G-) bacteria from type culture collections and clinical isolates, in planktonic and biofilm form. At first, the basic antimicrobial activity evaluation was performed by using the microdilution broth method. Additionally, the modified methodology was used to explore the influence of culture broth and initial bacterial density on evaluating the antimicrobial activity. Secondly, by using the quantitative suspension test, that better simulates normal practice conditions, has been successfully implemented at the department for the detailed antimicrobial activity evaluation. Rozpoznávání drah částic v pixelovém detektoru typu Timepix

In current particle physics field, the progressive detection technologies are used. The pixel detectors are one of them. These detectors are divided into small subdetectors (pixels), which allow viewing exact tracks of the detected particles. This thesis defines criteria for mathematical description of the shape of the particle tracks of different kinds (e-, γ, p, α, μ) and compares several methods used for a classification -neural networks, decision trees and others. The Pixa software was implemented to process the data measured by pixel detectors. This software implements the characteristics and classification methods and creates statistical and other physical results.

Their large system of conjugated bonds enables them to absorb light in the red part of the absorption spectrum. Due to their ability to produce singlet oxygen, they can be potentially used as photosensitizers in photodynamic therapy (PDT). Its mechanism is based on co-functioning of three elements - photosensitizer, light and oxygen. Photosensitizer excited by light absorption transfers its energy into tissue oxygen, thus, creating cytotoxic singlet oxygen. This method is beneficial for its high selectivity, low toxicity, minimal invasion and fast effect. The aim of this work was to synthetize and study water-soluble TPyPz suitable for PDT. Clostridium difficile: Molecular typing of clinically significant isolates

Currently, Clostridium difficile is a leading nosocomial pathogen due to the spread of epidemic strains. Molecular typing of clinical isolates is an important part of C. difficile occurrence and spread control in hospitals as well as in the community. A total of 2201 clinical C. difficile isolates from 32 hospitals cultured between 2013-2015 were characterized by PCR ribotyping and toxin gene multiplex PCR. A total of 166 different ribotyping profiles were identified, of which 53 ribotyping profiles were represented by at least two isolates for each profile. The most frequently found ribotypes were 176 (n=588, 26.7%) and 001 (n=456, 20.7%) followed by 014 (n=176, 8%), 012 (n=127, 5.8%), 017 (n=85, 3.9%) and 020 (n=68, 3.1%). Out of 2201 isolates, 2024 (92%) isolates were toxigenic and carried genes for toxin A and B, and of these, 677 (33.5%) also carried genes for binary toxin. The remaining 177 (8%) isolates were non-toxigenic. Subtyping of C. difficile isolates using a multilocus variable-number tandem repeats analysis (MLVA), that compared the sum of tandem repeats differences, was performed in C. difficile isolates of ribotype 176 (n=225, 17 hospitals) and in C. difficile isolates of ribotype 001 (n=184, 14 hospitals) cultured in 2014. Tuberculosis incidence differs considerably in various regions of the world but even the countries with low incidence engage strongly in epidemiology of tuberculosis. Tuberculosis belongs to one of the priorities of WHO, cooperation in this matter takes place on a global scale. Tuberculosis is a social illness; accordingly, the countries with high occurrence of tuberculosis are classified as developing countries. Mainly in Africa, there is the situation being complicated by coexistence of HIV. Generally, Europe represents a region with low incidence of tuberculosis. The methodology proposes criteria for determination of the content of micro-nutrients in soils and their evaluation via useing of the extraction method Mehlich 3. Extraction procedure Mehlich 3 is the official analytical method currently used in testing system of agricultural soils for the determination of the content of available P, K, Mg and Ca. There are criteria for valuation of content of these available nutrients - see Decree No. 275/1998 Coll., on Agrochemical Testing of Agricultural Soil and Land Surveys of Forest Lands, as amended). The existing criteria for above mentioned nutrients are due to this methodology extended by next five elements (Cu, Fe, Mn, Zn and B), marked in field of plant nutrition as micro-nutrients. The simple and easy Mehlich 3 analytical procedure applicable to a variety of different elements, is already used in a wide range of laboratories, allows to extend the criteria for the others elements. In addition, many laboratories can use the multi-elementar analytical instruments ICP-OES. Suggested criteria will also serve for farmers for valuation of soil supply by micro-elements.

Monitoring the effectiveness of marketing in the construction company

This thesis deals with monitoring the efficiency of marketing at construction company. The aim of the work is comparing the effectiveness of investments in marketing for the new and regular customers. The output of work is matching of the amount of investments made in marketing in contrast obtaining new and retaining existing customers during achieving the same profits.
