Utilisation of recycling machines in complex approach to waste treatment
Černý, Ondřej ; Máša, Vítězslav (referee) ; Bělohradský, Petr (advisor)
The purpose of the thesis is to describe and evaluate a contemporary state of waste management in the Czech Republic in comparison with European Union. One of its goals is to point out permanently increasing amount of waste, which is produced by people. It also analyzes the principle in the area of planning, future processing and utilization of waste material. Then it deals with the specific cases in the waste management system of entrepreneurial entity that was chosen by us, and with the machines including their technical parameters, which this entity disposes at present period with.
The costs of single phase of the building life cycle
Černý, Ondřej ; Vaňková, Lucie (referee) ; Krejza, Zdeněk (advisor)
The bachelor’s thesis determines the costs of individual phases of a building’s life cycle. The thesis explores a very broad subject which entails a variety of topics. The theoretical part concerns itself with building in general, costs of individual phases of a building´s life cycle and pricing in the Czech Republic. In its practical part, the thesis primarily dissects determining costs for the utility, maintenance, repair and reconstruction of a building during its life cycle. The process for the calculation of the total sum of financial costs in individual phases of the life cycle is shown in a table, which contains the costs for individual phases of the building’s life cycle.
Analysis and Design an Information System for Elegis s.r.o.
Černý, Ondřej ; Olexik, Martin (referee) ; Klusák, Aleš (advisor)
This work includes analysis of company processes, their demands on operational support by information system and analysis of current condition of information system. Based on this analysis, there are also several adjustments in processes and information system designed to make company work more efficient.
Assessment of the second viscosity coefficient from measured pressure pulsations.
Černý, Ondřej ; Pochylý, František (referee) ; Habán, Vladimír (advisor)
Diploma thesis deals with problem of assessment of the second viscosity and acoustic velocity coefficients by the different liquid´s flow in pipe from measured pressure pulstation created by diaphragm pulsator. For the solution is used a transfer matrix model and for the data evaluation a genetic algorithm in „Facharp.exe“ program. Modal analysis in problematic part of the measured stand is realized to find a wave shapes and natural frequency of a pipe. The results are for the reason of clearness processed into „MS Excel“ program and fetched to the charts which shows depedence of a second viscosity and acoustic velocity coefficients on a various flow as the final result.
Distribution system voltage stabilization by means of PV inverters control
Černý, Ondřej ; Novotný, Jan (referee) ; Drápela, Jiří (advisor)
This thesis is dealing with options for solar inverters control aiming to stabilize voltage in LV distribution networks. For this purpose, it evaluates the possibility of using reactive power control. Currently proposed methods of reactive power control are reviewed and a new method, based on combined reactive and active power control is proposed and tested.
Information System Assessment and Proposal for ICT Modification
Černý, Ondřej ; Hajn, Pavel (referee) ; Koch, Miloš (advisor)
The thesis is focused on an information system and management assessment of a company. This review is written up on the basis of analysis of the efficiency and the equitability of an individual information system's dimensions. Results of these analysis were later determined as a foundations for a practical part of the thesis which is focused on a suppression of identified issues of the information system and its management and on an enforcement of these improvements in the company.
Speed Measurement of Yacht
Černý, Ondřej ; Bradáč, Zdeněk (referee) ; Šedivá, Soňa (advisor)
Work deals with the measurement speed of yacht and wind speed. For measuring is used GPS module and cup anemometer. In the thesis is detailed analysis of the process and construction of eletrical equipment that processes incoming data of speeds and displayed on the screen. Thesis further processes the measurement results and compares the theoretical assumptions and practice
Czechoslovak newsreels from the perspective of social transformation (1989-1990)
Černý, Ondřej ; Lokšík, Martin (advisor) ; Moravec, Václav (referee)
This thesis deals with the end of film news in Czechoslovakia, which was realized in the form of film weekly newsreels until 1990. In the first part I define film news from the theoretical and terminological point of view and try to describe its specifics. I then explain its birth and the production of newsreel films on our country's territory, from its beginnings before the First World War until the fall of the communist regime in Czechoslovakia and the establishment of democracy, which ended the continuous, state- subsidized and controlled production of regularly published weeklies. In the second, practical part, I use the method of qualitative content analysis to examine the form and thematic framework of the weekly newsreels produced in 1989, when a new concept of film reporting focused on the so-called magazines began to take effect, and in 1990, the year Czechoslovakia entered the democratic process. My aim is to find out what was the role of weekly newsreels as a means of socialist propaganda and a channel of social criticism in the first, revolutionary year and how they changed in the second, democratic year. Although they were no longer bound by ideology, the end of the state film monopoly brought an end to their financial support and power protection, which led to an early end of their...
Factors contributing to stress adaptation of human pathogen Bordetella pertussis
Hejnarová, Václava ; Večerek, Branislav (advisor) ; Černý, Ondřej (referee)
The role of bivalent metals as cofactors in various enzymes is essential for all domains of life. Their importance can be seen in interactions of hosts with patogens such as Bor- detella pertussis. This bacterium has several mechanisms that protect it from oxidative stress in phagosomes of immune cells. One example is Mn-dependent enzyme superoxide dismutase. Phagosome itself is an environment with limiting concentrations of several key bivalent metals. Thus, the transport of these metals must be controlled to ensure sufficent concentration for enzymatic activity and prevent from toxic cumulation and mismetallation. This study focuses on a particular locus BP3083-BP3077 that is highly expressed in B. pertussis surviving after internalization by THP-1 macrophages. The locus consists of two transporters, one of which shares homology with SitABCD Mn2` impor- ter of Salmonella enterica and is regulated by Fur family repressor. Bacterial cells were exposed to toxic concentrations of several bivalent metals and the level of gene expres- sion was studied by RT-qPCR. Deletion mutants were used for examination of bacterial phenotype. Results of the study show that the locus encodes Mn2` and Co2` transporters and becomes negatively regulated by Fur family repressor in toxic concentration of Co2` . The...
Identification of residues of acylated domain of RTX toxins involved in acyltransferase binding
Grobarčíková, Michaela ; Mašín, Jiří (advisor) ; Černý, Ondřej (referee)
Both adenylate cyclase toxin (CyaA) and α-hemolysin (HlyA) are members of Repeats in ToXins (RTX) cytolysins that play key roles in the virulence of Bordetella pertussis and Escherichia coli, respectively. Bacterial RTX toxins represent a growing group of proteins produced by gram- negative bacteria. These pore-forming RTX toxins share several notable common features: (1) they require post-translational activation by attachment of fatty acid chains to two lysine residues; (2) they contain a hydrophobic domain that forms cation-selective pores in target cell membranes; (3) they are secreted by a type I secretion system; (4) after secretion, they become biologically active by binding of Ca2+ to the nonapeptide glycine- and aspartate-rich repeats. CyaA translocates a unique AC enzyme to the cytosol of phagocytes and subverts their bactericidal functions by unregulated conversion of ATP to cAMP. CyaA and HlyA also permeabilize the cell membrane of eukaryotic cells through cation-selective pores. Both toxins preferentially bind to cells expressing β2 integrins but can also interact with a variety of cells that do not express integrins or with naked lipid membranes. Both toxins are activated from protoxin form by post- translational acylation mediated by a specific acyltransferase. CyaA is activated by...
