The latest model of tertiary structure of capsid protein of potato virus X (PVX CP) was used as a template to design new insertion sites suitable for the preparation of PVX-based antigen presentation system. Based on this model, seven insertion sites (A-G) located in putative surface loops were tested. As an antigen inserted into these sites was used 17 amino acids long epitope derived from human papillomavirus type 16 E7 oncoprotein (E7 epitope) fused with either 6xHis tag or StrepII tag in both possible orientations (6xHis-E7 and E7-6xHis, StrepII-E7 and E7-StrepII). Prior to plant expression, modified PVX CPs were expressed in Escherichia coli MC1061. The results showed that only PVX CP carrying StrepII-E7 or E7-StrepII in the insertion site A formed virus particles. The results from transient expression experiments with modified PVX CPs in Nicotiana benthamiana showed that only the insertion site A (located between 24th and 25th amino acid in the PVX CP) could tolerate all tested inserts. Importantly, viral particles were detected only in the presence of StrepII tag and their stability was affected by the insert orientation (StrepII-E7 vs. E7-StrepII) as only the viral particles presenting E7-StrepII could be purified. Telemetry and Dispatch Control of Low Voltage Grid

The contents of this thesis are the introduction to mesh grids, the method of dispatch control of these grids and the description of technology used in mesh grids distribution nodes in the city of Brno. This thesis also describes low voltage switchgears used in these types of grids. The development of dispatch control in ECD company is also mentioned. The development describes the grid dispatch control methods prior to implementing the OMS system and the changes which followed after the implementation. The process improvements of the new system resulting from this thesis can be found in this thesis as well. The improvements are incorporated in the OMS system and are used for more efficient dispatch control of the low voltage mesh grids. There is a more detailed analysis of the mesh grid Brno – Bohunice in the practical part of this thesis. The practical part contains analyses of mesh grids stabilized conditions measurements and analyses of mesh grids fault conditions measurements. The analyses assess the power load in transformers, minimal and maximal phase current and maximal power load at the time they were observed. The analysis of the measured data is accompanied by the assessment of differences in current phase measurements and differences in voltage phase and combined measurements. The result, based on the analysis and collection of the data, is a proposition of adding switchgears for the support of the dispatch control of the mesh grid Brno – Bohunice. In the summary of the thesis there is a comparison of the result of the factual and theoretical analysis. An experimental model of the mesh grid Brno – Bohunice was created in PS CAD software as a part of the theoretical analysis.

Automatic Voltage Control of Distributed Generation in the E.ON Medium Voltage Grid

Content of this master`s thesis is theoretical introduction of the power factor, reactive power and voltage control of decentralised sources of the high voltage in the E.ON distribution network. Furthermore, the thesis deals with possibilities of regulating the power factor and reactive power, carried on above-mentioned resources, in other to stabilize the voltage and overflow control of the reactive power to higher voltage level. The practical part describes how the automatic power factor and voltage control had been put into action by central management control within the headquaters dispatching systém of the company. Following chapter acquaints a reader with information of how the automatic power factor and voltage control had been tested and how it is utilized in practice. In the conclusion the work summarizes results and effects of the power factor regulation and voltage control to stabilize the voltage and the overflow of the reactive power in the E.ON network.
Molecular-biological characterization of Potato virus M and Potato leafroll virus

The main aims of diploma thesis were: 1) The sequence analysis of the Czech isolate of Potato virus M (PVM) VIRUBRA 4/009 and phylogenetic analysis of PVM coat proteins sequences 2) The bacterial expression of recombinant triple gene block protein 1 (TGB1) of PVM derived from the Czech isolate VIRUBRA 4/007 3) The construction of expression cassette of Potato leafroll virus (PLRV) coat protein and its transformation into A. tumefaciens for transgenic PLRV resistant plant formation In theoretical part of the thesis the taxonomic classification, morphology, genomic structure and virus transmission are discussed. Furthermore, the main rules concerning the bacterial expression of recombinant proteins and construction of transgenic plants using A. tumefaciens are described. Methodical part is devoted to description of generally used molecular biological and immunochemical methods.
