The role of aquaporins in the Alzheimer's disease

Alzheimer's disease (AD) is a progressive neurodegenerative disorder with complex pathophysiology affecting the central nervous system (CNS). In progress of the disease, various pathological changes occur in the brain, leading to neurodegeneration and subsequent impairment of physiological and cognitive functions. Although it is the most common cause of dementia in elderly, currently, there is no effective treatment for AD that that targets its underlying mechanisms. There are different theories as to which process is the key trigger for the development of AD. The generally accepted theory considers increased production of amyloid β (Aβ), its accumulation in the ECS and the formation of amyloid plaques as the main cause of the disease. However, recent studies show that the primary cause of amyloid plaque formation is not increased Aβ production, but rather its impaired clearance through the glymphatic system, the main component of which are aquaporin water channels, specifically aquaporin-4 (AQP4). The goal of this thesis is to provide an overview of the available knowledge on the involvement of aquaporins in AD pathophysiology, with a particular focus on AQP4 and its role in the glymphatic system. Identification of changes in membrane properties of astrocytes in a mouse model of amyotrophic lateral sclerosis

Amyotrophic lateral sclerosis (ALS) is a progressive neurological disorder of the central nervous system characterized by loss of motor neurons and voluntary muscle degeneration. Astrocytes play a major role in regulation of the disease onset and progression due to their intimate association with neurons. Regulation of ionic homeostasis is one of their key functions and its failure has been linked to several neurological diseases. The aim of this thesis was to explore differences in membrane properties of astrocytes in ALS. To fulfill this aim, a double transgenic mouse strain with ALS-like phenotype and a specific expression of enhanced green fluorescent protein in astrocytes was generated. To phenotype this strain, two sensorimotor tests, wire grid hang test and rotarod test, were conducted. Immunohistochemistry was used to characterize the strain on a cellular level and to explore changes of specific ion channels. Functional properties of astrocytes were explored using the patch clamp technique. The double transgenic strain has the characteristic ALS-like phenotype and is comparable to the original strain with differences in symptom onset and progression between models and sexes. Tato diplomová práce pojednává o nové formě obchodní korporace , jejíž zavedení na vnitřní trh Evropské unie by mělo usnadnit společnostní v zahraničí, a to zejména pro malé a střední podniky. Návrh směrnice společnostech s ručením omezeným s jediným společníkem, který přináší formu vnitrostátní obchodní korporace, která se bude řídit harmonizovanými pravidly napříč všemi členskými současné době předmětem legislativního procesu v Radě EU Potřeba představení této nové formy obchodní společnosti společně s absencí české odborné literatury k tématu byly primárními Kromě samotné analýzy návrhu směrnice jako celku a jejich jednotlivých ustanovení původního návrhu předloženého Komisí a kompromisního znění návrhu, jehož bylo dosaženo při projednávání směrnic prvním čtení v Radě v obecného přístupu, a jejich kvalitativní zhodnocení. Použitými pracovními metodami jsou především metoda systematické analýzy, deskripce a komparace. Vyjma úvodu a závěru se tato diplomová práce skládá ze tří kapitol dále členěných zájmu přehlednosti předložení návrhu směrnice o společnostech s ručením omezeným s jediným společníkem, čímž vytváří podklad pro pochopení cílů a důvodů tohoto návrhu. ormy obchodní společnosti, přičemž směrnice znění předloženém Evropskou komisí. The Control of Private Military and Security Companies

The thesis deals with the regulation of activities of private military and security companies in armed conflicts. The aim of the thesis is to outline the current state of mechanism of control concerning the participation of private military and security companies in these conflicts in order to achieve compliance with international humanitarian law and human rights law. The first and the second chapter characterize armed conflicts and the phenomenon of their privatization from the theoretical point of view. The third chapter discusses the mechanism of control on the international level which is based on the Montreux Document. The last chapter describes a self-regulation mechanism in the private security industry, its structure and functionning. The thesis emphasizes the role of the International Code of Conduct for Private Security Service Providers and the ICoC Association, the first oversight body of its kind.
