Mass transfer during desorption

This master thesis deals with development and optimalization of apparatus for ammonia desorption from liquid organic wastes, generated by biogas plant. There is a high ammonia concentration in biogas plant waste products. That limits its application as a fertilizer in an agriculture. The main aim of this theses is finding optimal conditions to reach required efficacy of removing ammonia nitrogen from a liquid digestate. For this purpose, two pilot plant devices were used, and optimal conditions for ammonia desorption with its subsequent absorption by acid solution were investigated. One of the tested devices is a membrane contactor with polymeric hollow fibers. The second is a stripping device, which was used for measuring the pilot scale experiments. At the time of writing of this thesis, the device is subjected to a process of optimalization. The transferred ammonia nitrogen absorbed in acid solution was analyzed by UV-VIS spectrometry. The suitability of both devices for the required ammonia desorption efficiency has been demonstrated. Furthermore, the kinetic curve of the experiment was investigated under different conditions. The optimalization of devices will take place according to the results obtained in the experimental part of this thesis. The optimized devices will be placed in biogas plant full operation.
Determination of content of selected nonsteroidal antiphlogistic – Ibuprofen in drugs using UV/Vis and FTIR spectrometry

The main aim of bachelor thesis was to validate the content of the nonsteroidal antiphlodstic in drugs using spectrometric methods and subsequent comparison of results. Specifically, the thesis is focused on Ibuprofen, which is used mainly as analgesic and antipyretic. This substance was chosen for its significant ability of absorbing electromagnetic radiation in ultraviolet and infrared regions of spectra. For this purpose, various generics were selected and analyzed by molecular absorption spectrometry (UV/Vis) and infrared spectrometry with Fourier transform (FTIR). Very important factor influencing chemometric properties of the obtained results was optimalization of the spectrometric methods. After designing the working procedure, sample solutions were prepared from Ibuprofen-containing dosage forms using suitable solvent and these samples were analyzed spectrometrically. The content of Ibuprofen in individual samples was calculated from the measured UV/Vis and FTIR spectra using method of standard addition. The results show that infrared spectrometry seems to be more suitable method for determining the Ibuprofen content. Nevertheless, it should be noted that the choice of a suitable extractant is very important step in the determination. The information obtained from this bachlor’s thesis could be used in future routine inspections and monitoring of drugs.
The role of regulatory protein SGIP1 in nociceptive synaptic transmission at the spinal cord level.

Cannabinoid receptor 1 (CB1), abundantly expressed in the CNS, is a promising target for the pharmacological treatment of pathological pain conditions due to its function as an inhibitor of neurotransmitter release from presynaptic neurons. Recently, the SGIP1 protein has been found to interact with the CB1 receptor and participates in the modulation of nociception. However, whether SGIP1 modulates spinal CB1 receptor signaling at the spinal cord level is unknown. To answer this question, we used the patch-clamp method in the superficial spinal cord dorsal horn neurons of wild-type (WT) and knockout (KO) SGIP1 mice to measure spontaneous (s) and miniature (m) excitatory postsynaptic currents (EPSCs). Results of naive mice and mice with carrageenan-induced peripheral inflammation were compared. The results show that the efficacy of the CB1 receptor agonist WIN 55,212-2 at the first spinal nociceptive synapse is identical in naive mice for both SGIP1 WT and KO phenotypes. The control frequencies of both groups of neurons did not differ in naïve conditions or the peripheral inflammation model. On the contrary, the WIN 55,212-2 application was more effective in SGIP1 KO mice during peripheral inflammation.
