Background: The aim of this diploma thesis has been to present an invertebrate organism, Galleria mellonella (G. mellonella), Pyralidae, as an alternative, more economically advantegous and morally more acceptable model for in vivo studies of an antituberculotics (anti-TB) efficacy. In the experimental part of this work, attention was paid to the optimization of methodological steps, that should lead to a higher reproducibility and validity of the obtained data. The effect of the disaggregating supplement, tyloxapol, on the growth of Mycobacterium aurum (M. aurum) was evaluated. The growth of this microbial strain in the culture medium, Middlebrook 7H9, was also mapped to determine the exponential growth phase. In the second part of the experiments, G. mellonella larvae were infected with a M. aurum bacterial strain, followed by an administration of the selected, commercially available anti-TB. A proposal of production technology of a rotary part

The theoretical part is focused on the theoretical description of the used machining technologies. The practical part is focused on the analysis of the current method of production selected component and design of a new method of production, the selection of a suitable machine and tools. Last part of this thesis is economic evaluation.
Animation on the Periphery

The thesis attempts to precisely describe the production of animation in the context of the state- socialist mode of production. Through the question of "autonomy" for cartoons and puppets films in the era of Kamil Pixa (1969-1985), we will try to specify the position of animation within the re- centralized organization and dramaturgical section in the Short Film. In the context of the normalization period, animation had a different position and function compared to the feature films. Using the example of a studio in Čiklova Street, we outline a model of making a puppet film that was more like a "craft workshop" that retained its notional autonomy as long as it had its own space.
Optimizing of rotating components production

The subject of this bachelor thesis is the analysis and following optimalization of technological process of manufacturing the component „Pouzdro“ which is turned on CNC lathe Hardinge Talent 8/52 and milled on Kovosvit MAS MCV 750. The goal of this bachelor thesis is design of production method when will used only CNC lathe DOOSAN PUMA 2100SY, choice of the tools and comparsion of this two methods of the technological processes with regard to production time, lifetime of the tools and economy of production.
Handicap as a Gift

In my thesis the potential of technical struggles and various mechanical limitations of puppets themselves is explored. Related to that also other limitation of puppetry and even drama as a genre itself are uncovered. Surprising analogy with the world of people with disabilities can be found here. So a disabilitity becomes a substantial point of view and a kind of subjects of demonstration which have been used in my work up to now. In the first part resources and terms related to disabilities are considered as well as the reasons triggering my interest in this phenomenon. A particular project subconsciously drew my attention to my higher sensitivity to various forms of disabilities in life and art as well
SPECIFICS OF CREATING SOUND DESIGN OF ANIMATED FILM

The thesis explores the production of sound for animated film – its specifics as well as aspects of realization and of preparation and audio dramaturgy. It examines how they impact storytelling and the extent to which it is possible and useful to work with them from the very beginning of development of an animated film. The thesis also presents the practical and creative possibilities of cooperation between director and sound designer, alongside factors that may have a positive or negative influence in this regard. It draws on literary sources, interviews with the makers of animated films and knowledge acquired by the author in the field of directing and screenwriting of animated films. The thesis confirms the importance of the role of sound in animated film and its positive influence on the resulting quality of AV work (unless it has been omitted from the start of the filmmaking process). It summarizes the basic approaches that allow animators to work with sound in animated film as a fully-fledged narrative component.

Bachelor thesis Overcoats presents unrealized projects (mostly films) of seven czech major directors of animated films. Text contains samples from specific projects, accompanying text of author of the thesis, which puts works into larger context and interviews which was made directly with authors of projects or the individuals most connected. Interviews were taken, rewritten and modified in July and August 2016.
Robinson Crusoe - comparison of animated movie and live action movie

The topic of this bachelor thesis is comparison between screenwriting for animated film and live action film and a search for differences in both types of writing. Specifically, this thesis compares two films based on the book The Life and Strange Surprising Adventures of Robinson Crusoe of York, Mariner, by Daniel Defoe. Using this book I am able to describe the possibilities of both literature and film, and to demonstrate which parts of the book are preferred for film adaptation and why. I also briefly mention the evolution of the story of Robinson Crusoe, and show how animation and live action films deal with these changes. In the final part of my thesis I compare the 1954 live action film Robinson Crusoe, directed byLuis Bunuel with the 1981 Czech puppet film The adventure of Robinson Crusoe of York, mariner, directed by Stanislav Látal. Comparing these two films enabled me to understand and describe specific rules and procedures in scriptwriting for an animated film.
Politics, Animation and Comics

Korean cinematography gained liberty after the military regime, but the situation depends on political situation. The growth of Korean cinematography shows influences of the policies by every korean government. Thanks to the world-wide success of Korean films, korean viewers perceive how the conservative government tries to control the film industry, and they know how to react to it. For example, the film 26 years (2012) - it was not possible to be shot because of the interruption by the conservative government. Because this film contains massacre in 1980 in the city Kwangju by the dictator, and the dictator is still alive. However future viewers of the film supported it, and the production invited subscriptions to a fund. Finally the film was finished in 2012, and distributed to cinemas all around south Korea. However korean animation is out of viewer's concern, even though it has been the biggest scapegoat by policies of Korean government. Every conservative government have used korean comics as a scapegoat whenever they reached problems. When there were serious criminals of youth or criminals against children, korean government strengthened censorships about comics. Some comics were banned, and authors of these comics could not work on their comics. It happened in 1997, and has began again in 2012.
