Sublingual drug delivery. 6
Janečka, Josef ; Doležal, Pavel (advisor) ; Vachek, Josef (referee)
Work in theoretical work deals with physiological barriers sublingual application of particle carriers, notes the results of authors who have dealt with similar themes. In the experimental part was to evaluate the permeability of membranes used sublingual modified Franz diffusion cells for horizontally positioned membrane dividing the chamber of the donor and acceptor part. For the research were used frozen pork sublingual membranes. Acceptor phase was isotonic phosphate buffer pH 7.4. Donor phase was 0.5% solution of fluorescently labelled particles PD Chromeon 470. To determine the concentration of particles in the acceptor phase was used spectrofluorometer Aminco Bowman Series 2 from Thermo Fisher. To detect low concentrations was used the highest sensitivity of the device and multiple measurement of samples. The average flux value of fluorescently labelled particles Chromeon PD 470 40 nm in size in five chambers measured during the first 12 hours of 0,18 μg.cm-2 .hod-1 reached.
