Use of cell cultures to test cytotoxicity of natural substances

This thesis is aimed on study of cell cultures, primarily on introduction of techniques for cultivation of human cells and optimization of their usage in cytotoxicity tests. The thesis is aimed preferably on natural substances and liposomes that are suitable to be used in cosmetics. Theoretical part contains literature review focused on overview of cell cultures, appropriate cultivation conditions for in vitro testing and summary of cytotoxicity tests. Experimental part contains characteristics of natural extracts with antimicrobial effect in terms of content of polyphenols, flavonoids and antioxidant activity. Further, these extracts were encapsulated into the liposomes. Encapsulation effectivity was determined. Distribution of particles size by dynamic light scattering and finally also stability of prepared particles were also determined. Long-term stability in water and gel environment was tested too. This thesis also contains study of optimization of cell culture of human skin keratinocytes cultivation and use of those cells for testing cytotoxicity of selected natural substances by MTT method.
The Perception of Poverty by Second Grade Pupils at Selected Elementary School

The diploma thesis focuses on the perception of poverty among the pupils of the second grade of the selected primary school - Primary School and Kindergarten Dolní Poustevna. Specifically, it deals with the extent to which pupils distinguish subjective perception of poverty, whether they are able to distinguish groups of people at risk of poverty, whether they are oriented in the possible assistance to people at risk of poverty. Distinguish and describe different aspects of child poverty. Interpretation of the issue. The theoretical part is divided into two parts. The first part explains and describes aspects of poverty, from the definition of poverty itself, to its concepts, causes, measurement, the poverty cycle and the issue of social exclusion. The second part focuses on the issue of the child and his/her needs. It deals with the psychological and biological development of the child and the importance of the family in the child's life. Attention is also paid to the social and legal protection of the child, the prevention of poverty and the role of the state in this issue. Ways to spend free time of primary school teachers in Skalica district

The diploma thesis examines the immediate leisure time of elementary school teachers working in the Skalica district, responding to the current situation where the issue of teacher burnout is increasingly acknowledged in both pedagogical and broader public discussions. Utilizing the acquired theoretical knowledge, the author characterizes free time from a historical perspective, identifying specific periods in which she observes disturbances in the utilization of leisure. The author will describe the changes reflected in the ways of spending free time nowadays The historical context enhances our understanding of the development and evolution of leisure and its content, both in the past and present, with clearly defined functions. A separate chapter delves into the description of leisure functions, providing insight into their significance. The author focuses specifically on the possibilities for spending leisure time in the Skalicky district. Finally, the theoretical part introduces us to teachers, outlining their duties and various ways of spending their leisure time. Lecturer of suburban camps

The bachelor's thesis entitled "Lecturer of suburban camps" deals with the professional prerequisites that a lecturer of suburban camps should have. The theoretical part of the thesis explains the professional areas of free time pedagogy, which are closely related to the teaching activity. It also deals with basic legislation that concerns teaching staff and suburban camps. The aim of the bachelor's thesis is to compare the requirements of the lecturers and the demands of the organizers for a new lecturer and, based on this, to propose the structure of the selection process and the content of the refresher course for lecturers with non-pedagogical education. The output of the research part of the work is to describe the professional requirements of the organizers for potential lecturers based on interviews. I also deal with pedagogical education and the experience of lecturers. At the end of the work, I present the recommendations resulting from the interviews for the organizers of suburban camps. Leisure activities of second degree primary school pupils from socially disadvantaged background

The bachelor's thesis on the topic Leisure activities of children from socially disadvantaged backgrounds focuses on how children from socially disadvantaged backgrounds spend their free time. Thesis objective is finding out what activities the second grade students from this environment choose and if and how often they use the free time activity offerings at school or nearby institutions. Last but not least, find out if the free time aktivity offerings of school and surrounding institutions are sufficient, or what other free time activities the pupils would be interested in. The thesis is divided into two chapters, a theoretical and a practical part. The theoretical part defines the concepts of free time, it's functions and the importance of leisure time education. It also deals with social disadvantage and identifying pupils from a socially disadvantaged environment, which was key for the practical part and choosing the research respondents. The second chapter describes the research group and the evaluation of acquired research data. The research part contains the results of a questionnaire survey, describing how children from socially disadvantaged environment spend their free time, which institutions these children visit and what leisure activities they prefer. This Bachelor thesis is dealing with the use of research-oriented teaching during hobbies as part of leisure activities in the field of natural sciences, especially chemistry. Nowadays is noticeable trend to start with research activities and building relation to natural and technical sciences at very young age of students. In the theoretical part the thesis describe principles and forms of research-oriented teaching and their realization, lecturer qualification conditions and work safety during the experiments in the hobby, development of students potential during younger school age, specifics of this students period and characterizes leisure activities in detail and their delineation in relation to formal education. Part of the practical part is a local survey of the accessibility of research-oriented clubs in the Polepy and Libochovice districts.
