Surrounding structures including proteins may be damaged in case the content of a phagocyte is spilled to the vicinity. Catechins belong to a group of flavonoids whose antioxidant properties were demonstrated in many studies. My diploma thesis was focused on a potential effect of eight naturally occurring derivatives of catechin towards oxidative damage of human serum albumin (HSA) by hypochlorous acid. I used two spectrophotometric (determination of carbonyl groups and chloramines) and three electrophoretic methods (native PAGE, SDS-PAGE, immunoblotting). Dependence of an antioxidant effect on its concentration (0-3 mM) was observed during determination of chloramines and obtained data were used for calculation of EC50 values of individual catechins using program GraphPad Prism. The most effective compound was catechin whose EC50 was 4.8 µM. Results suggest that antioxidant ability decreases with increasing number of hydroxyl groups in the molecule of flavonoid. The role of PML nuclear bodies in herpesvirus infection

PML nuclear bodies are protein structures in the cell nucleus that regulate many important cellular processes and are also implicated in antiviral defense. The permanent components of these nuclear bodies include PML, Sp100 and Daxx proteins, many other proteins are transiently associated with PML bodies. PML body components can be SUMOylated, this posttranslational modification is important for the formation of PML bodies and regulation of their function. Components of PML bodies such as PML, Sp100 and Daxx can act as restriction factors limiting the replication of many DNA and RNA viruses. Defense mechanisms mediated by PML bodies are suppressed by viral proteins that inactivate individual components or disrupt the structure of PML bodies. This thesis focuses on the role of PML bodies as restriction factors during infection by DNA viruses of the Herpesviridae family and describes the interactions of PML bodies and viral proteins, using herpes simplex virus 1, human cytomegalovirus and Epstein-Barr virus as examples. Design of Helium liquefaction cycle

The present master thesis deals with the technologies for gas liquefaction focused on helium. The first part of the thesis contains a description of the history of liquefaction, production and usage of liquid helium, as well as the principles of liquefaction and cooling cycles. In the following part of the thesis is a detailed model of the liquefaction cycle for which radial-axial turbine wheels were designed. The last part of the thesis contains financial analysis of the suitability of using the expander with eddy current brake and the expander with an electric generator.
Source for heat production in the Czech Republic

This bachelor thesis deals with thermal recources in the Czech Republic. The first part of the work is about the division and description of heating plants used for the production of thermal and electric energy. The second part of the thesis describes the advantages and disadvantages of district heating and individual heating. The last part of my work contains statistical processing of heat sources located in the Czech Republic.

Analysis of Motivation System of State Land Office Employees

The goal of my diploma thesis is to analyze and summarize general knowledge of work motivation and its application in the environment of the State Land Office (hereinafter SLO). A part of the goal is also my own suggestion of precaution and recommendation, how to improve satisfaction of employees and the ability of the middle and the higher management to motivate subordinate employees. To fulfil the goal a method of quantitative research has been used, in which we work with all the SLO employees. Evaluation of the data in relation to the set hypothesis was done in form of calculating averages of respondents answers and at the same time comparison of satisfaction and importance of work satisfaction factors. The diploma thesis has come to conclusion, that the SLO employees are satisfied with some of the other work satisfaction factors, than those which are important for them. A part of the conclusion was also a discovery, that the employees have many suggestions and impulses to make changes, which should take place within the framework of the SLO. Results of the questionnaire contributed to launching of a new SLO project, which deals with improvement of work satisfaction of the SLO employees.
Development Assistance in Post-conflict Reconstruction in Africa: Case Study of Rwanda and Burundi

The thesis deals with development cooperation in post-conflict reconstruction. The comparative case study analyzes the specific involvement of the international community in the post-war reconstruction of Rwanda and Burundi with emphasis on two main areas, political/constitutional, and socio-economic. Based on the findings of the analysis, possible measures are suggested to make development cooperation in post-conflict reconstruction more effective, especially in the case of Burundi. The results of the analysis show that the international community should in case of these two countries, among other things, mostly focus on securing political stability, supporting civil society, or building infrastructure, and thus help to achieve the goals of post-conflict reconstruction.
