Detekce variability cpDNA u rostlin

This thesis deals with the detection of the variability of a given region of chloroplast DNA. A selected region of chloroplast DNA matK is compared between two genera Aloe and Ruschia as representatives of the two evolutionary lineages of monocots and higher dicots. CpDNA is widely used for phylogenetic studies due to high variability. Selected 92 representatives from the genus Aloe and 106 representatives from the genus Ruschia were used for analysis. Both genera are predominantly found in southern Africa, mainly in the Cape region. The first part of the thesis was focused on the characteristics of the selected genera and the description of the methods used in the analysis. The analysis involved isolation of DNA from representatives of both genera, PCR, purification and subsequent sequencing. The obtained sequences were evaluated using available software and subsequently phylogenetic trees of the selected representatives of both genera were constructed. The phylogenetic trees were created using the Maximum likelihood method. The main result was the detection of variability within and between the selected genera based on consensus sequences.

The ITS1-5,8S-ITS2 sequence region of ribosomal DNA represents one of the most popular molecular marker in phylogenetics. However, detailed analyses of 5,8S rDNA are scarce and most of them deal with the complete region including the internal transcribed spacers ITS. In this thesis, 5,8S rDNA sequences were subjected to bioinformatic analysis to compare the variability of ribosomal genes in monocot and eudicot plants. The analyzed sequence data of the regions ITS1-5,8S rDNA-ITS2 were obtained by an experimental approach from isolated DNA and by searching in nucleic acid databases. The variability was assessed in total 114 sequences for each group. Multiple sequence alignments were used to identify conserved regions and to quantify gene mutations. Higher heterogeneity was detected in the 5,8S rDNA sequences of the eudicot group, but no statistically significant difference in the representation of variable sites (15,2 % and 19,3 %) or in the total number of mutations was confirmed. Manual pseudogene detection, based on plant conserved 5,8S rDNA motifs search, sequence length, content of GC and in silico secondary structure prediction, revealed a total of 12 potentially non-functional copies of the 5,8S rDNA gene.
Variabilita polymorfizmu v genu FABP3 a jeho asociace s ukazateli kvality masa u prasat

The aim of the study was to study the variability of the gene in the population FABP3 pig breed Czech large white. Analyze their association with indicators of pork production. Gene FABP3 belonging to multigene families FABP (fatty acid binding proteins affecting the transport of long chain fatty acids). Analyzed polymorphism was at position X98558:g.1321G>C in the 5'UTR region. The relative frequencies of genotypes were: HH = 0,87, Hh = 0,13. hh genotype was not detected in the monitored individuals. The values of the relative frequencies of alleles were as follows: H = 0,93 h = 0,07. Based on the association analysis showed a statistically significant difference between genotypes for stearic acid and linoleic acid. Highly statistically significant difference was observed with palmitic acid.
Long-term tendencies of spatiotemporal differentiation of unemployment in Czechia with emphasis on seasonality

This master thesis belongs thematically to the research of long-term trends of spatio-temporal differentiation in the Czech Republic. Within this theme, it focuses on several objectives. The first of them concerns the search for appropriate scale levels and region types for time-space analyzes of unemployment in the Czech Republic and the definition of as many homogeneous regions as possible in terms of the unemployment rate. Subsequently, the work at the appropriate level defined in this way focuses on the next goal, which is to reveal and describe the development of seasonality in unemployment in the Czech Republic. As part of this goal, the effort is to determine the significance of the phenomenon of seasonality in unemployment, its development in space and time, and to define groups of regions that are more or less affected by this phenomenon. One of the partial goals is also to reveal the development of spatial differentiation in unemployment in the last few years. The work uses detailed monthly data on the unemployment rate in the Czech Republic at the municipal level for the years 2002-2021 published by the Ministry of the Interior. Auxiliary selection in present-day Italian

This thesis is dedicated to researching the use and importance of auxiliary verbs in tempi composti (compound tenses) in Italian language and grammar. Some of these verbs show variability and in some instances they might prefer a different auxiliary verb than is the one commonly allocated to them. In the theoretical section of this thesis, compound tenses and auxiliary verbs are introduced. That is followed by the explanation of theories that were presented by authors Antonella Sorace and Delia Bently, including non-accusative hypothesis. The research section of this thesis was based on following the categories presented in the non-accusative hypothesis. Numerous various verbs were chosen and subsequently tested in CORIS and ItWac. Their occurrence and preferences towards the auxiliary verb(s) was compared and contrasted with the above mentioned thoeries.

Title: Applied pharmacokinetics in preclinical research Author: Mgr. Jana Královičová Abstract: This thesis addresses the topic of development and practical utilization of rat models in preclinical pharmacokinetic studies. An analysis dealing with the impact of intra- and inter- individual variability on the results of animal comparative studies showed that cross-over design brings more accurate conclusions in these experiments than the often-used parallel setting. Results obtained on the basis of a parallel design significantly differed in our investigation (AUClast ranged from 9.62 to 44.62 mg/ml.min.g). Furthermore, in 4 cases out of 15 pair comparisons of individual parallel groups, the confidence interval did not include the 100% value. That means that in more than a quarter of studies it would be misjudged that the compared drug formulations have different pharmacokinetic properties, though they are in fact the same product. The practical use of cross-over design is demonstrated in pharmacokinetic studies comparing the absorption of various formulations with abiraterone and cinacalcet. In a fasted state, a new, innovated formulation with abiraterone was better absorbed than the reference (AUClast of a reference 30.3 mg/ml.min.g, innovated formulation 81.0 mg/ml.min.g). The main purpose of this thesis was to find out, whether and to what extent the rate of dispensing, the dispending angle (90ř, 65ř, 45ř) and the volume of preparation (30 ml, 15 ml, 5 ml) had effected the mass of commercial eye drops for poultry vaccination. Drops were produced manually by using conventional dispensing system (plastic 30 ml dropper bottle and dropper tip) and by using a new prototype of semi-automatic device. The drop mass increased with increasing the rate of drop formation in every examined combination of factors. With decreasing of the dispensing angle, the drop mass decreased only in a case of constant rate dropping. In all other cases, especially with volume reduction to 15 ml and 5 ml, the drop size often increased due to the wetting of hemispherical dropper tip. As a result, higher variability of drop masses had occurred. With using the semi-automated prototype, the preparation volume had no impact on the final drop mass. On the other hand, higher variability of doses was noted for studied suspension eye drops.

In this thesis, the mass of veterinary eye drops for chicken vaccination was evaluated. Drops were produced either by conventional dosing system or by new developed dropper device (semi-automatic). The influence of dosing rate, dispensing angle (90ř, 65ř, 45ř) and volume of liquid (5 - 30 ml) on the mass of eye drop was investigated. The higher dosing rate resulted in the increase in drop mass. The influence of dosing rate on the mass of drop was significant (p < 0.01) for both dispensing systems. The drop mass decreased with the decrease in the dispensing angle using manual dosing system if the eye dropper tip was not wetted, with the wetted one, the drop mass increased in comparison to vertical dropping. Due to the construction of the dropper device, the influence of dispensing angle was not evaluated, however, the wetting of dropper tip was not observed thanks to defined perimeter. The reduction in the remaining preparation volume led to the decrease in drop weight, particularly with simultaneous dispensing angle decrease, the effect was not significant.

The bachelor thesis deals with the design of a residential building in an almost neglected location in Moravská Ostrava. The proposal looks for a way how to portray housing as such as community housing applicable in Czech conditions. The result is an adaptable environment that users can choose according to their wishes, as democracy says - about choices, about voting. Its variable and flexible form offers very easily and quickly changes to the program, whatever the advanced age of users, or with the needs for more space for new additions to households. Even the smallest apartments take on a sense of generosity, with pull-out furniture and wall - screens. The interior of the flats is renewed and creats a "room in a room". However, the case study also deals with the housing crisis and the shortage of housing, so the variety of functions are limited mainly to housing. However, the capacity for variation are retained and can be adapted over time to community housing, mixed - used or office space in an attractive location. It addresses the housing crisis comprehensively, and with a choice of apartment sizes ranging from studio apartments to 3 bedroom apartments. This system can be applicable to a variety of areas as well as plots with numerous dimensions. One of the many goals is to connect the public space with the indoor and outdoor space of the building. But it is it is important to maintain the intimacy of not only the semi-public space - the common areas of the building, but also the intimacy of the apartments. The design becomes a meeting point between the city and nature, specifically the roof garden and root purifier located in an unenclosed atrium in the wider centre of Ostrava. At the same time, it is capable of lowering the temperature in a densely populated and sparsely vegetated area.
Democratic Architecture – Kamenný vrch

The bachelor thesis deals with architectural design of a building near gable of a prefabricated house on Kamenný vrch in Brno. My building is one of four designed projects at this spot. All the buildings created the center that has been missing before. It brings new functions to the locality and also new people. Residents of the building are constantly changing thanks to its function, which are offices for startups, temporary accomodation and added foyer with small bar not just for residents. Space is the most variable as it can be, so it can be used by residents as they please.
