The effect of aspartate β-hydroxylase inhibition on immune reactions

Aspartate β-hydroxylase (ASPH) is an enzyme that contributes to tumor progression by enhancing the proliferation, migration and invasiveness of cancer cells. Its expression is mostly negligible in normal cells, whereas it is often overexpressed in cancer cells. An inhibitory effect of ASPH on immune cells, specifically on natural killer cells, has also been demonstrated. Thus, the ASPH enzyme could be a new target for cancer immunotherapy. The aim of this thesis was to show the effect of ASPH inhibition on the immune response. Firstly, it was found that ASPH inhibition contributes to the anti-tumor effect of DNA vaccination. The reduction of tumors induced by ASPH inhibition in combination with DNA vaccination was shown to be mainly caused by CD8+ T lymphocytes. Subsequently, the specific activation of T lymphocytes was confirmed by the ELISPOT assay. In case of non-specific activation of T lymphocytes, ASPH inhibition had no effect on the direct activation of CD8+ T lymphocytes. Finally, it was found that plasmacytoid dendritic cells did not contribute to CD8+ T lymphocyte activation after ASPH inhibition. Thus, the results demonstrate that inhibition of ASPH contributes to the activation of adaptive immunity induced by DNA vaccination, but the mechanism of this activation remains unknown.
Role of Zinc-α2-glycoprotein in tumors

The Zinc-α2-glycoprotein (ZAG) encoded by the AZGP1 gene particularly contributes to the regulation of lipid and glucose metabolism. In different types of tumors, its altered production was found and the association of the ZAG level with the patient's prognosis was suggested. However, the role of ZAG in tumor progression can differ in individual malignancy types and mechanisms of ZAG function are not sufficiently identified. The objectives of the bachelor's thesis are a description of the structure of the ZAG protein and the regulation of AZGP1 expression, a summary of the identified ZAG functions in nonmalignant diseases, and a focus on the role of ZAG in tumors, particularly on the mechanisms of its action and its potential use as a biomarker.
Role of NAV3 in glioblastoma cells invasiveness

The invasion of tumor cells from the primary lesion and the formation of metastases are the main reasons for the severe impact of cancer diseases. An option for dealing with this poor impact is the development of drugs (so-called migrastatics) that would target these processes and thus limit the spread of tumor cells from the site of the primary tumor. However, to develop such drugs, it is essential to clarify the molecular mechanisms that control or promote cell migration. One of the possible strategies for migrastatics development is the targeting of cytoskeletal structures, which play an indispensable role in cell migration. This work is focused on Neuron navigator 3 (NAV3), a protein that binds to + ends of microtubules (MTs), participates in their stabilization, and is able to mediate crosstalk between MTs and the actin network. The function of NAV3 is important for directing MTs into growing axons and proper neurite outgrowth, which is necessary for brain development. The results of this thesis suggest that NAV3 could act as a pro-tumor factor, which localizes not only to the + ends of MTs, but also to the cell protrusions, and whose presence supports the cell expansion and increases the invasive potential of glioblastoma cell lines. Oncological diseases in the Czech Republic, the second leading cause of death. Tumors of the digestive tract in this balance occupy an important place. The least favorable prognosis have tumors of the pancreas, liver, stomach, esophagus and intestines. The Bachelor thesis deals with a brief description of the phenolic substances and antioxidant component in wine and also introduces the reader in the context of the relation of wine and antioxidant components of selected gastrointestinal tract authorities, on the basis of the clinical studies that were studied. In conclusion, the thesis is a recommendation, which is evaluated from the processing of individual studies.

Anti-tumor activity of mesenchymal stem cells

Mesenchymal stem cells (MSCs) are multipotent cells with the ability to migrate to inflammation sites and to tumor sites. They are able to regenerate the damaged tissues and also easy to isolate and cultivate. Furthermore, they can inhibit tumor cells and modulate the immune response. They are non-toxic in the organism and genetic modification of them can enhance their antitumor effect. MSCs can also serve as a vehicle for delivery of the therapeutic agent to the tumor. These properties make them special for anti-tumor therapy. Under some conditions, MSCs can also stimulate the tumor growth. This work discusses conditions in which MSCs inhibit the growth of cancer cells, as it is not yet clear on which precise mechanisms this inhibition is based.
Iron metabolism in cancer cells

1Abstract: Cancer is one of the major causes of death in the present world. As the research of this disease has progressed, the attention of some scientists has been focused on a metabolism of iron and how it can be used to fight these rapidly proliferating invasive cells and stop their spreading. This work should serve as a brief review of iron metabolic processes from the iron absorption from dietary resources and recycled cell iron, to its usage in heme- or Fe/S clusters-proteins and storage in a form of ferritin, while highlighting the points that differ in cancer cells. It also gives a modest overview on the regulatory pathways of iron uptake and use, and mentions iron metabolism disorders such as iron-depletion and overload. Simultaneously it is denoting possible differences that could be targeted in tumor treatment, and, at least but not last, the perspectives and future work that could bring a new methods and approaches to this matter. Correlation of Immunohistochemistry and Molecular Biologic Methods in The Diagnosis of Tumors

This thesis is a collection of commentaries on altogether 13 first-authored and 20 co- authored publications where morphology, immunohistochemistry (IHC) and molecular genetic methods were used to provide novel clues for arriving at an accurate diagnosis of tumors, as well as to propose novel approaches and refinement of classification of certain tumors. The presented manuscripts are the result of the postgraduate studies of MUDr. Michael Michal at the Charles University in Prague, Faculty of Medicine in Pilsen in the period between 2015-2018. The author focused the main part of his research, particularly his first-authored manuscripts, on soft tissue tumors but also largely participated in research activities focusing on other body systems. Over the course of his studies, four main areas of interests within the topic of soft tissue pathology emerged. The first is oriented on soft tissue tumors of presumed (but unconfirmed) fibroblastic lineage. First two publications regard two related low-grade sarcomas called Myxoinflammatory fibroblastic sarcoma (MIFS) and Pleomorphic hyalinizing angiectatic tumor (PHAT). The role of mitochondrial respiratory chain in invasiveness and metastasis of cancer cells and possible therapeutic interventions

The mitochondrial respiratory chain, also called the electron transport chain (ETC), has a pivotal role in key features of cancer cells e.g., proliferation, the metabolic shift from oxidative phosphorylation (OXPHOS) to aerobic glycolysis, or the ability to form metastases. This review summarizes current knowledge about ETC and its relationship to cancer, especially to invasiveness and metastases formation. Firstly, it deals with a process called the Warburg effect and with metabolic complexity in the tumor microenvironment. Then it shows how OXPHOS activity affects invasiveness of cancer cells and metastases formation, and it points out the connection between invasiveness and increased levels of ETC-generated reactive oxygen species. At the end, the review deals with possible use of ETC inhibitors in anticancer therapy.
Development of total mortality and mortality from selected causes of death in the Czech and Slovak regions since 2001

The main aim of the bachelor thesis is to analyse total mortality and selected causes of death in Czechia and Slovakia, based at regional level. It means a comparison of their development for each region in both countries. The analyse is monitored from 2001 to 2018. The first part of the analysis is devoted to the analysis of the total mortality using various indicators as a life expectancy at birth and life expectancy at 65 years. Also, this part is finished by decomposition method which complements the results of life expectancy at birth. Following the development of total mortality, second part of the analysis is about selected causes of death. Only the diseases that are most numerous in both countries were chosen for this part of the thesis. In terms of total mortality, the main result is that mortality rates are improving in the Czechia and Slovakia, which is reflected in increasing of life expectancy. As part of the analysis of mortality from selected causes of death, it was found that during the observed period there was a decrease in the intensity of mortality from the category of diseases of the circulatory system. The trend from mortality in cancer is slightly different, because while the intensity of mortality is declining in the Czechia, in Slovakia the trend is very slow for men and essentially...
