Construction and evaluation of in vitro set of holes for chondrogenic differentiation of MSC cells

Bachelor thesis concentrates on the cultivation of mesenchymal cells in vitro, damage to the articular cartilage and its replacement with the chondrogenic cells. The next section is devoted to each type of temperature control for heating and maintaining the temperature inside a closed system, the type of detector temperature and heating using the Peltier element. Assembly heater box, measuring the temperature profile inside the box with Labview program, control with Arduino platform, survival and differentiation of cells.

Studium faktorů ovlivňujících mikropropagaci oskeruše (Sorbus domestica L.)

This diploma thesis was elaborated on the theme "The study of factors influencing service tree (Sorbus domestica L.) micropropagation". The experiments were carried out in in vitro laboratory of the Mendeleum - Institute of Genetics and Plant Breeding of the Faculty of Horticulture in Lednice. The aim of this work was to acquire and improve the knowledge of in vitro cultivation of serviceberry and the genus Sorbus L. in general. The trees growing in experimental land of the Faculty of Horticulture MZLU in Lednice were selected as experimental material. The composition of cultivation medium was verified for plant growth and multiplication purposes. The influence of two different cytokinins BA (benzyl-adenine) and zeatin was evaluated. The shoot number and shoot length were measured. The highest multiplication rate evaluated by the origin of new formed shoots was accomplished on media supplemented with BA (0.1 mg.l-1). Nevertheless the shortest height of shoots was measured on this media. It was valid for both cytokinins that number of shoots was increasing with increasing cytokinin concentration whilst the shoot height was decreasing.

In vitro reproduction represents a vegetative way of multiplication, in which small disinfected pieces of plant (explants) are cultivated in a nutritious medium in sterile conditions in order to gain whole new plants. This method, also called micropropagation, is often used for research purposes and more and more often for commercial applications too. This work studies nutritious media for apricots. For Prunus armeniaca cultivars best described media are for bud and embryo cultures. Many different media were tested and it was shown that every cultivar of apricot has its own requirements of nutrition and growth regulators. In this work you can find descriptions of many successfuly used media for apricots along with responses of various cultivars to them.
Cultivation of grapevine in conditions in vitro

Grapevine is a traditional crop in the Czech Republic, especially in the South Moravia region. It is a vegetatively propagated crop; hence vineyards are endangered by abiotic and biotic stress influence. The risk of plant damage due to frost and pest influence occurs in the field conditions according to season and locality. Cultivation of grapevine in conditions in vitro eliminates the risk. Higher utilization of the method is limited by sensitivity of some genotype to in vitro conditions. This methodology solves these problems and describes material introduction into in vitro conditions, explants multiplication and slow growth condition establishment. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

The issue of transfer selected hybrids of the orchids genera, grown under aseptic conditions in vitro, into non-sterile conditions

In this paper experiments were made with three taxa of orchids. The first part was to find out whether the treatment of plants with potassium permanganate (KMnO4) has any impact on mortality during the transfer from in vitro culture to the unsterile environment of routine cultivation. All three taxa were divided into two parts and only one part was always treated while the other not. The deaths in individual groups were recorded during the experiment. In the second part the effect of fertilization on the biomass growth was investigated on two taxa. The method was again based on the division of the taxa into two parts and the fertilization of only one part. Necessary data were obtained by the weighing of plants at the beginning and end of the experiment. The results of the first experiment indicate that treatment of plants during the transfer has no impact on the subsequent mortality. In the second part, the dependence of fertilization on the biomass growth has been successfully proven in only one of the two taxa.
