The subject of my thesis was an analysis that deals with the level of nutrition of holstein cattle, witch evaluates feeding, drinking, silage quality techniques and optimalization of the feed ration in 2017 and 2018. In 2017 the feed ration used for dairy cows contained more PDIE. In 2018 the feed ration contained lower amount of energy (6MJ NEL/ 1kg of dry matter) and NL (138,6 g NL/kg of dry matter). After comparing of these two diets for dairy cows with nutrient need, there was recommended an increase of dry matter content in feeding ration. In the milk productivity assessement, the fat and protein content, SC, total aerobic microbial count, was reviewed. The statistic shows the impact of the season of the year on the fat and protein content, SC, total aerobic microbial count in milk. From ecomomy point of view, costs per litre of selled milk was 7,69 Kč in 2017, which was lowered to 7,28 CZK per litre of selled milk. Both years was profitable, and in comparing the economy from 2017 with Czech republic average, tracked stable was more profitable 3 662 CZK per dairy cow.

This thesis purpose is to evaluate level of nutrition of dairy cows Holstein cattle on the farm. Backround research was made for this topic. Claim of the study is to compare nutritional value of the feed rations with the recommended nutrient requirement ford dairy cattle. The feed ration for cows meet recommended nutrient requirements. Feed rations for the dry-off cows contain high level of nitrogenous compounds. The correction was recommanded. Further, the quality of silage roughage was assessed. Compared milk yield was lower by 615 kg milk/cow/year from the performance of Holstein cattle population in the Czech republic.

Under operational conditions the concept of nutrition and efficiency of milk cows (the breed: Holstein Friesian cattle) was assessed in the chosen agricultural enterprise during the years 2015 and 2016, as well as the production of feed for the milk cows and the economical aspect of milk production during the period of decreasing prices of its purchase. The thesis embodies the assessment of the negative impact of a low rainfall total in the locality of monitored enterprise specialising in the production of bulky feed. Because of this fact the feed amount of productive milk cows were changed in 2016. An interannual decrease of milk yield about 1,92 l per 1 peace of kettle and 1 day was recorded on the grounds of the change of feed doses. The decrease in efficiency and consequently the decrease in delivery of market milk to trade in 2016, together with low prices of purchase of milk, had direct influence on a low economical profits in the area of milk production and milk-cows breeding of the monitored enterprise.

The diploma thesis analyses the level of nutrition of dairy cattle of a particular company and looks at factors that influences milk production. Under the operating conditions of this particular company is evaluated: the quality of bulk feed, the composition of feed diet in terms of required amounts of nutrients and energy for production levels and compared the recommended values.The thesis assesses feeding technique, housing technology and hygiene of obtaining milk. The production and health of dairy cattle breeding over the course of one year is depicted. The work also focuses on evaluating the basic economic indicators of milk production.
Analysis of cow milk caseins by capillary electrophoresis

The diploma thesis is dealing with the determination of caseins in lyophilised samples of milk using the method of capillary electrophoresis (CE). This method was optimised and verified by Ivana Micikova in her diploma thesis in 2009. The theoretical part of the diploma thesis gives information about the chemical compound of cow's milk. Caseins and their usage in the food and non-food industries are described in a separate unit. Impact of both the nutritional and non-nutritional factors on the composition of the milk is discussed further. The description and the principles of the CE and a list of the methods of CE used in the last several years are stated at the end of this part. The experimental part is focused on the preparation of the solutions, process of casein isolation and its analysis. It summarises the setting of the experiment, sampling and sample processing and the composition of the feeding mixtures. The diploma thesis has arisen on the basis of the cooperation of the Institute of Food Science and Biotechnology, the Faculty of Chemistry, Brno University of Technology and the Research Institute for Cattle Breeding, Ltd., Department of Animal Nutrition Physiology in Pohorelice.
Milk composition changes after fortification milch cow feed with selected additive

This thesis is dealing with effect of supplement limiting aminoacid histidin, that was applied as duodenal infusion, on yield and composition of high production dairy cows milk. The theoretical part describes overview about milk composition and metabolism of dairy cows. There are described nutrition requirements of dairy cows for esential aminoacids. In this part are also presented principles of analytical methods used for analysis of milk. For determination of basic components of milk were used infrared spectroscopy and enzymatic-conductometry. Aminoacid profile of casein was determined by ion exchange liquid chromatography. In the experimental part is described method of experiment. There is described technique of acid and oxidative hydrolysis of lyofilized casein and technique of determination aminoacids in this hydrolyzated casein. Results of analysis were interpreted and discussed with literature. This diploma thesis was made as a cooperation between the Institute of Food Science and Biotechnology, the Faculty of Chemistry, University of Technology, Brno and the Research Institute for Cattle Breeding, Department of Animal Physiology and Nutrition, Pohorelice.
Determination of caseins by capillary electrophoresis

The diploma thesis, Determination of the caseins by the capillary electrophoresis, is dealing with a creating of the analytical method for a determination of the caseins in cow‘s milk. The capillary electrophoresis was used as a separative method. The developed procedure was optimalized and verified on standards of the caseins and the real samples. The theoretical part gives an information about the compound of cow's milk, caseins, their usage in the food industry and the non-food technologies. There are mentioned some further nutritional factors, whose application in the cattle nutrition can affect the milk composition. The experimental part describes the procedure of the caseins isolation from milk, the used separative system and the determination of the caseins in the lyophilisated samples of milk. The separation was performed in two untreated fused-silica capillaries. Both of them had the same internal diameter: 50 m. The total length of the first capillary was 96 cm (71 cm of effective length) and of the second used capillary 125 cm (effective length 100 cm) The phosphate buffer (10 mM, pH 2,5) served as a basic electrolyte. The diploma thesis has arisen on the basis of the cooperation of the Institute of Food Science and Biotechnology, the Faculty of Chemistry, Brno University of Technology and the Research Institute for Cattle Breeding, Ltd., Department of Animal Nutrition Physiology in Pohořelice.
Determination of caseins in cow milk

The diploma thesis is dealing with design of analytical method for determination of caseins in cow‘s milk. Capillary electrophoresis was used as separative method. The developed procedure was verified on real samples. Theoretical part gives basic information about caseins. Their chemical and physical properties, structure, way of manufacturing, usage in food industry and non-food applications are described. There are further some nutritional factors mentioned, whose application in cattle nutrition can affect milk composition, especially milk protein concentration. Literary part contains summary of capillary electrophoresis methods used for casein determination. Experimental part describes used separative system, procedure of casein isolation from milk and casein content in individual milk samples. Separation of milk caseins was performed in an untreated fused-silica capillary with dimensions of 50 um i.d. x 96 cm total length (71 cm effective length). Phosphate buffer (10 mM, pH 2,5) served as the background electrolyte. Casein from milk was obtained by isoelectric precipitation at pH 4,6 after adding 10% acetic acid. Product was washed with acetone to remove fat and dried. Total casein yield was the highest in bio-milk (27,04 g/l), the lowest in skim long-life milk (22,04 g/l). Analysis of the operating indicators of milk production

This thesis analyzes the operating characteristics of milk production. The work was developed in agricultural cooperatives of Zemědělské družstvo Podkleťan Křemže. The agricultural holding had been in assessment of dairy cattle nutrition, feeding techniques, quality of roughages, composition of the diet and comparison of economic indicators in dairy production. The quality of roughages was performed by using the fermentation process, and the fodder was subsequently classified into different classes. The composition of the diet was assessed with recommended values depending on the energy need and nutrient requirements. The reproductive performance was compared with data on total population. The economic factors were assessed in connection with yield and cost of farming.
