Mouse mammary tumor virus (MMTV) is a prototypical member of the Betaretrovirus genus characterized by the ability to preassemble viral particles in the cytoplasm of the host cells. Intracellularly preassembled particles are subsequently transported to the plasma membrane being enveloped by a lipid bilayer and released from the cell in the process referred to as budding. Retrovirus particle assembly is driven by the Gag polyprotein precursor, which is cleaved in the maturation process by virus-encoded protease to liberate multiple structural proteins. The matrix (MA), capsid (CA) and nucleocapsid (NC) protein domains that are common to all retroviruses and in the case of MMTV, also the noncanonical domains, pp21, p3, p8 and "n", located between MA and CA domain are present. The role of these specific domains remains undefined. The retroviral budding is stimulated by short peptide motifs, so-called late (L) domains, located within Gag sequence. These L domains mediate interactions with cellular proteins normally involved in the biogenesis of the multivesicular bodies and protein sorting. Vliv proteinu HBx viru hepatitidy B na aktivaci MEK1/2-ERK signalizace a inhibici IFN typu I v hepatocelulární linii Huh7

Infekce virem hepatitidy B (HBV) je jednou z hlavních příčin chronického a nádorového onemocnění jater. Eliminace HBV z chronicky infikovaných pacientů monoterapií založené na rekombinantním interferonu α (IFNα) ukazuje, že mechanizmy přirozené imunity hrají důležitou roli při potlačení virové infekce. Mechanismy rozpoznávání genomu HBV a jeho úniku před mechanismy přirozené imunity jsou však stále málo známy. Jedním z principiálních faktorů umožňujících viru uniknout před buněčnými restrikčními mechanizmy je virový protein HBx. HBx je pleiotropní multifunkční protein složený ze 154 aminokyselin ovlivňující transkripci, přenos signálu, buněčný cyklus, degradaci proteinů, apoptózu a chromozomální stabilitu v hostitelské buňce. Předchozí výsledky z naší laboratoře ukázaly, že aktivace signální dráhy MEK1/2-ERK v plasmacytoidních dendritických buňkách vede k inhibici produkce IFNα. Cílem mojí práce bylo zjistit, zdali HBx aktivuje dráhu MEK1/2-ERK a inhibuje tím produkci IFN typu I rovněž v hepatocytech. K tomu účelu jsem sledovala tvorbu HBx v hepatomové linii Huh7 pomocí transfekce bicistronického plasmidu pHBx-IRES-EGFP a metody Western blot. Stejnou metodou jsem sledovala aktivaci signální dráhy MEK1/2-ERK pomocí fosforylace kinázy ERK. Moje výsledky ukazují, že maximum exprese HBx v buňkách Huh7...
