Vaňková, Radka ; Struhala, Karel (referee) ; Košíčková, Ivana (advisor)
This bachelor’s thesis deals with the processing of project documentation for the implementation of construction, semi-basement, detached house with hairdressers´s. The building is designed slightly sloping terrain. The hairdressers´s is situated in the front part of builfding with entrance from east side. It is soluted like a ground-floor part of object with wheelchair access. In this part of building is a garage too. It´s connected on the residental part of building by the vestibul. The entrance to the residental part of the building is from the north side. The resident part is made from ground floor, second floor and partly from the basement. In he basement, there is a technical facilities. In the ground floor there is a living room together with a kitchen. The another rooms are on the second floor.
Analysis of Sensitization to Molecular Components Determined by the Multiplex System Immunocap ISAC in Patients with Atopic Dermatitis
Vaňková, Radka ; Čelakovská, Jarmila (advisor) ; Nečas, Miroslav (referee) ; Stříž, Ilja (referee)
Background: Atopic dermatitis (AD) is a chronic inflammatory skin disease with acute exacerbations and characteristic clinical manifestations. The diagnosis of AD is based on the medical history, evaluation of clinical condition, supplemented by skin or exposure tests. An integral part is also the examination of specific IgE antibodies against extract allergen or single molecular component. Biochip multiplexing techniques have recently been used for respiratory forms of allergic inflammation and when food allergy is suspected. However, these analyses could find application in the diagnosis of cutaneous manifestations of allergic inflammation, such as AD. The aim of this work was to more acurately identify the source of food and inhalant allergy in patients with AD. We evaluated sensitization profiles in patients with mild, moderate, and severe forms of AD and in a subset of patients with AD complicated by bronchial asthma and allergic rhinitis. We also addressed the sensitization to molecular components in the context of food allergy in patients with AD. Methods: Blood serum samples were taken from adult patients with AD. Levels of specific IgE antibodies were determined by the ImmunoCAP ISAC multiplex system. The dissertation thesis summarises already published results of our scientific team....

Charles University Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Radka Vaňková Supervisor: doc. PharmDr. Martin Beránek, Ph.D. Title of diploma thesis: Influence of Carrier Molecule Addition on Plasma Free Circulating Nucleic Acid Extraction Outline: The aim of this thesis was to compare the yields of cell-free DNA (cfDNA) extracted from blood plasma specimens with the addition of commercially available carrier molecules. According to the real-time polymerase chain reaction (qPCR) results, we chose the most optimal carrier molecules for routine isolation of cfDNA from blood plasma samples. Methods: cfDNA was isolated from aliquots of a pooled blood plasma by spin column extraction method NucleoSpin Plasma XS (Macherey-Nagel). We used 7 different types of carrier molecules as RNA carrier (Qiagen), Glycogen (Invitrogen), Poly (A) Polyadenylic acid (Roche), Linear Acrylamide (Invitrogen), Yeast transfer RNA (Ambion), Salmon sperm DNA (Invitrogen), Herring sperm DNA (Promega). After the extraction, plasmatic DNA with the addition of carrier molecules was quantified by spectrophotometry, fluorimetry and qPCR. Results: The best results were achieved with the addition of polyadenylic acid with final concentration of 1.55 μg/ml. The addition of carrier RNA and...
Hyperviscosity Syndrome in Patiens with Monoclonal Gammopathy.
Vaňková, Radka ; Fátorová, Ilona (advisor) ; Vrbacký, Filip (referee)
The thesis deals with the assessment of correlations between selected laboratory parameters used in the diagnosis hyperviscosity syndrome and the determination of monoclonal immunoglobulin. By means of specific examples is graphically illustrated the effect of therapeutic plasmapheresis on laboratory parameters and the clinical condition of the patient. The thesis is divided into theoretical and practical parts. In the theoretical part are summarized the findings of the diagnosis, clinical manifestations and therapeutic methods of hyperviscosity syndrome. Further the history of monoclonal gammopathies is stated with splitting in two basic groups. Within the group of malignant monoclonal gammopathies are described in detail two diseases that are closely related to hyperviscosity syndrome. The first is Waldenström's macroglobulinemia and the second is multiple myeloma. In both diseases is shown their prevalence, definition, etiology, pathophysiology, laboratory findings and clinical symptoms. Further are briefly described methods used for determination of monoclonal immunoglobulin. The practical part describes the laboratory tests, which were obtained laboratory data of the reference group of patients. Laboratory data were processed by regression in Microsoft Office Excel 2007. A statistically...
Analysis of chosen factors influencing the decision about educational level in the Czech Republic
Vaňková, Radka ; Bolcha, Peter (advisor) ; Bydžovský, Jiří (referee)
The graduates of universities in most cases will have better future than the graduates of lower levels of education not only in terms of higher wages, but also social status in broader sense. In spite of this not all graduates of secondary education choose to continue in tertiary education. That is why this bachelor work deals with chosen factors, that could have the impact on the individual's choice about tertiary educational level. Theoretical part mainly stands upon the publications and papers of Gary S. Becker and Jacob Mincer, on the human capital analysis and also on the documents, annual reports and data of OECD and Czech Statistical Office. For purposes of empirical check we use the data from the Czech Republic. To answer our question we use the method of regression analysis and the test of hypothesis about equality of two mean values. The goal of this paper was to find out if and how the chosen variables influence the individual's decision to graduate the tertiary education mentioned above, and to interpret measured outcomes in the context of the relevant theories. In the Czech Republic setting we have not found evidence that exogenous variables have significant impact on endogenous variable (even thought in some foreign papers this impact was evidence). However some outcomes led to interesting findings and showed possible factors entering to human decisions.
