This thesis is focused on accidents in water and their their division into groups according to different factors. Work detailing the chemical accidents resulting in pollution of surface and groundwater, which became the Czech Republic since 2000. Describes chemicals that often escape into the water and damage to their quality. Furthermore, there is described an accident in 2006, when form "Lučební závody Draslovka, Kolín" leaked into the Elbe dangerous cyanide. Key words: Accidents, water pollution
Development of on-line SPE HPLC method for determination of ochratoxin A in Tokaj wines

RNDr. Dalibor Šatinský, Ph.D. Title of Diploma Thesis: Development of on-line SPE HPLC method for determination of ochratoxin A in Tokaj wines This diploma thesis deals with the optimization and validation of chromatographic method for determination of ochratoxin A in Tokaj wine with subsequent application on real samples. A high- performance liquid chromatography method with fluorescence detection using on-line extraction by column switching has been developed. 50µl of sample was injected directly in the extraction column, where at 1st minute the analyte was extracted on the Ascentis® Express RP-Amide precolumn in the presence of a mobile phase of 40% MeOH in 0.5% aqueous acetic acid. After the extraction, the valve was switched and the extracted substances were separated on a YMC Triart C18 ExRS analytical column. The separation was carried out under gradient elution with an initial acetonitrile ratio of 45% in the mobile phase, which reached a concentration of 80% in the 5th minute. Evaluation of the quality of water and sediments in the basin Kunratice stream

This thesis deals with the monitoring of water quality and aquatic sediments analysis in the basin of Kunratický brook. Water samples were collected at six locations during once year. The results of analysis were evaluated according to CSN 75 7221 Water quality - Classification of surface water quality and Regulation 229/2007. According to CSN 75 7221 Kunratický brook is classified as very strong polluted water. Biological consumption of oxygen, ammoniated and nitrate nitrogen meet not the conditions of environmental quality according to Regulation 229/2007. Sediment samples were collected once for whole period at 4 locations. In sediment was determine the content of metals. The results of analysis were evaluated according to Regulation 257/2009. The results were compared with limits of US EPA, specifically PEC and TEC. Sediments of pond Šeberák are full obliged. Zinc, lead and cooper are in excess of sediments of Kunratický brook.
