Analýza metabolitů v přírodních povrchových vodách

Annotation: Natural waters contain dissolved organic carbon (DOC) in various extent. It is com-posed of metabolites, organic compounds involved in cellular metabolism. Amino acids, sug-ars and organic acid are the main representatives that occur at very low concentrations in these waters. The main goal of the thesis was to develop a method for direct determination of selected metabolites in natural fresh surface waters by liquid chromatography coupled to mass spectrometry (LC-MS). For the selected metabolites, the method was validated. In the next step their concentration was determined in water samples obtained from the regional surface waters. Amino acids phenylalanine and lysine represent major detected metabolites that were observed in the concentration range between 0.5 and 25 ng.ml-1.
Size-exclusion chromatography - analytická metoda pro charakterizaci rozpuštěných organických látek

The main aim of this bachelor thesis is the application of size-exclusion chromatography for a measurement of the molar mass of organic matter in water samples from Bohemian Forest lakes and subsequent determination of iron, aluminium and phophorus con-centration by inductively-coupled plasma/mass spectrometry. In the theoretical part, there is information about organic matter in water and the principles of size-exclusion chromatography and ICP/MS. The experimental part deals with the procedure of organic matter measurement and the determination of iron, aluminium and phosphorus concentration. Afterwards, the procedure of SEC and ICP/MS data processing is described and the results are discussed and compared with other analytical techniques.
