The influence of acyclic nucleotide phosphonates PMEG and PMEDAP on p38 kinase signaling in human leukemic cells

PMEG [9-(2-phosphonomethoxyethyl)guanine] and PMEDAP [9-phosphonomethoxy- ethyl)-2,6-diaminopurine] are acyclic nucleoside phosphonates possessing cytotoxic properties. Antiproliferative effect of PMEG was demonstrated in various tumor cell lines in vitro. PMEG also represents an active component of some experimental prodrugs with enhanced selectivity and efficacy (such as GS-9219). PMEDAP seems to have weaker effect in vitro compared to PMEG, however it exhibited pronounced antitumor effect in SD-rats with spontaneous lymphoma. Therefore it was included in the present study as well. The aim of this study was to describe the interactions of PMEG and PMEDAP with p38 MAP kinase signaling and its relationship to the apoptosis. We investigated the influence of these compounds on the expression of four genes encoding p38 MAPK isoforms and whether this change is translated into the protein. It was found that PMEG up-regulates p38β and γ mRNA in CCRF-CEM cells and p38 β and δ in HL-60 cells. The effect of PMEDAP was less pronounced than that of PMEG. However, total p38 protein level remained unaffected by PMEG and PMEDAP. Activation of p38 MAPK cascade was also measured in the cells exposed to these agents using phospho-specific antibodies. The use of alternative metods of pain relief in labour

This Baccalaureat thesis discusses the use of alternative methods of pain relief in labour. The theoretical section describes labour pains and the physical process of giving birth. This is followed by a number of alternative methods suitable for use during the birth, how they work, the possibilities for use, indications and contraindications, and ways of using them. Broadly speaking, the empirical section was completed using methods of quantitative research observation. It should clarify which alterative methods were offered to the labouring women and which were used. The study also looks at how women generally deal with pain, who helped them most during labour and whether the experience of giving birth met their expectations. The respondents were drawn from families using four medical institutions: Hradec Kralove University Hospital, Liberec regional hospital and Hořovice hospital. The 4th institution, a university hospital, wishes to remain anonymous. Women with planned ceasarean sections were excluded from the study.
Economical effectivity of growing of selected crops in organic and conventional agriculture

The diploma thesis compares economical effectiveness of selected crop cultivation (namely winter wheat and oats) in conventional and ecological economic systems during the years 2007 - 2014. Assessment of the economical balance was done through detected data about yield and redemption price. In addition to that, costs of both area and production unit within both mentioned types of farming were determined according to standard technological approaches. The study further deals with crop yields differences analysis within individual farming systems, the costs structure and price differentiations between ecological and conventional production. It also covers the question of the amount of subsidies and its influence on the final crop profitability.
The health issues of the Staphylococcus genus

The bachelor thesis deals with the information primarily regarding the diseases caused species of genus Staphylococcus and problems increasing of resistance to antibiotics. The disease of mastitis of dairy cattle belongs to the most serious and costly laborious facts in the agriculture companies. In the bachelor thesis are analysed alimentary intoxications in humans caused by staphylococcal enterotoxins. The antibiotic resistance is growing all over the word and the inception of the resistance is much faster than the discovering of new antibiotics.
