Myxosporean phylogeny and evolution of myxospore morphotypes

Evolution of the Myxozoa, the group of extremely reduced metazoan parasites, has been studied on the molecular phylogenetic level for more than a decade. This thesis is focused on morphological and molecular characterization of myxozoan species with emphasis on genera with missing molecular data in GenBank as well as on revealing the hidden and cryptic species myxozoan diversity. Evolution of specific traits in myxospore morphotypes and estimation of the time of the myxozoan divergence is also investigated and further discussed.
Roles of CD4+ and CD8+ T-lymphocytes in immune response to the gastric cryptosporidiosis

The roles of CD4+ and CD8+ T-cells were studied by using Cryptosporidium muris and mouse model of infection: immunized and naive BALB/c mice as a source of cells for adoptive transfer of immunity to SCID mice. Naive and primed CD4+ T-cells have a major effect in immunity of cryptosporidial infection. However SCID mice reconstituted with primed CD8+ T-cells were able to recover from the infection. The purity of the adoptive transfer pre and post-inoculation is discussed.
Occurrence of cryptosporidia in exotic mammals kept in zoological gardens

A total of 331 faecal samples of 102 exotic mammalian species were collected in four zoological gardens in the Czech Republic and examined for Cryptosporidium spp. oocysts by staining method, immunofluorescent assay for detection of cryptosporidia and PCR of the SSU rRNA loci. In this study, the prevalence of these parasites was found to be low. Cryptosporidium sp. was found only in a reticulated giraffe (Giraffa camelopardalis reticulata). Sequence analysis identified our isolate with C. muris genotype previously found in rock hyrax (Procavia capensis) and bactrian camel (Camelus bactrianus). Our Cryptosporidium isolate was not infectious for neonatal and adult mice BALB/c.
