Surface-enhanced Raman spectral detection of bilirubin and temporally synchronized monitoring of its photochemical transformations in selected solvents by SERS and electronic absorption spectroscopy
Hrnčířová, Jana ; Vlčková, Blanka (advisor) ; Procházka, Marek (referee)
Surface-enhanced Raman scattering (SERS)-active systems based on macroscopic Ag nanosponge aggregates as well as the conditions of SERS spectral measurements were optimized for selective and sensitive detection of a biomedically important, amphiphilic bile pigment bilirubin (BR) in alkaline aqueous solutions and in its solutions in a selected water miscible solvent, namely dimethylsulfoxide (DMSO) and/or water-immiscible solvent, namely CH2Cl2. Ag nanosponges assembled by using HCl as a pre-aggregation agent were found to be the optimal SERS-active systems for a reliable detection of BR in all the above-mentioned solvents. In all cases, the protonated form of adsorbed BR has been detected upon BR incorporation into Ag nanosponges, and its marker bands have been established by SERS spectral probing at excitation wavelengths in the 445-780 nm range. The sensitivity of SERS spectral detection was evaluated in terms of the concentration values of SERS spectral detection limits (SERS LODs) of BR incorporated into Ag nanosponges. In particular, the SERS LOD for BR incorporated from its alkaline aqueous solution is 1 x 10-8 M (at 532 nm excitation), for BR incorporated from its solution in DMSO, its value is also 1 x 10-8 M (at 532 and/or 633 nm excitations), and for incorporation from the solution of BR...
Origin of biomolecules over clays under conditions of early terrestrial planets
Hrnčířová, Jana ; Ferus, Martin (advisor) ; Vaculovičová, Markéta (referee)
The mankind has always been interested in the origin of life on Earth or in the universe. The Oparin's theory1 replaced creationist metaphysical theories (of gods' actions on Earth to create life) in the beginning of the twentieth century. This theory explained production of basic building blocks of life like amino acids, nucleobases and sugars from abiogenic mixture of gases and subsequent formation of so-called coacervates. The synthesis of amino acids was then confirmed by experiments of Miller and Urey2 . Especially the theory of simple formation of nucleobases became an essential contradiction between evolutionary and molecular biologists and chemists. From biological point of view it seems very probable that primal life structures were based on the self-replicating molecule of RNA. From chemical point of view the synthesis of nucleobases seemed very problematic, therefore the models of life structures based on amino acids were favorited. This opposes the basic dogma of molecular biology, which postulates one-way transcriptions of nucleobases' sequence into amino acids' sequence exclusively. In our experiments we follow the idea of simple production of nucleobases from formamide molecule in possible primordial conditions. The mixture of formamide with different types of clays was exposed to...

Executive search
Hrnčířová, Jana ; Šikýř, Martin (advisor) ; Matula, Miloš (referee)
In the theoretical part of the thesis focuses on the general definition of the recruitment and selection, mainly the methods, which are used. Furthermore, it defines the area work in public administration with a focus on employment and workers. Also, try to determine the difference between the acquisition employees in the public and private sectors. The work is then engaged in recruitment agencies and HR consulting companies, their history, the legislative definition and the question whether it is advantageous to work with recruitment agencies. Afterwards, the method of work is engaged in executive search, its history, use, and especially how it is itself the recruitment method. The practical part contains specific case of HR consulting company that deals with executive search method. Here is then analyzed in the procedure of recruitment executive search method and success by getting employees the company achieved in 2011. This part of the thesis is based on long-term observations of the author, who was an employee of the staffing and consulting company so he could observe the whole process of executive search methods in practice. Using a questionnaire survey, this work investigates how executive search method is used in the private and public sectors and what are the respondents as the main advantages and disadvantages of the method. At the end of this section for recommendations on how to use executive search method in the public sector, and problems are defined here, why not in the sector executive search method does not use too much.
Convention of the rights of the child and its consequence for pedagogical theory and practice
HRNČÍŘOVÁ KONSTANTINOVÁ, Jana
This thesis is covering the Convention of the rights of the child, its consequences and use in pedagogical theory and practice according to the paragraph 42 of this convention. It has three main parts, which are aiming for following the logical consequence of the topic compilation. First part is covering theoretical base of the Convention, starting with its historical development and current state, legal position, institutional arrangement, approaches to the Convention by selected European countries, including analysis of individual articles and it also presents commentary to these articles in connection with legal regulations in Czech Republic and finding the causes and deficiencies of its implementation, observing to it and its fulfilling. Second part is covering the definition of article 42 of the Convention as to the further education of adults as a tool for exercising the rights of children and to other theoretical and practical possibilities of its further efficient dissemination. Third part is related to that as it provides practical exercises, directions and games, how to get children acquainted with the document of the Convention not only during the school, but also within their free-time activities, including the chance for children to take part in the children{\crq}s parliaments or establishing own school self-governments and thus participate on public matters.
Social security for people living within European union for example
Hrnčířová, Jana ; Ševčíková, Michaela (advisor) ; Durdisová, Jaroslava (referee)
The Bachelor thesis comprehends how to coordinate the social security system within the European Union, that it isn't subject to unnecessary barriers of free movement. The first part will mapp of social security from the Middle Ages to the present and further work aims to coordinate, its principles and means of implementing the various components of social security and coordination in the course of practice. The second part will be devoted to specific social security systems in selected countries. Countries, whose social welfare work will be addressed, are Belgium and the Czech Republic. States were selected based on their position in the EU, where Belgium represents one of the founding members, while the Czech Republic is cited as an example of new accession countries. The aim of this work is closer cooperation tendencies of the system between the EU and through the comparison of selected countries to determine the extent to which different countries social security systems changed.
