Bachelor Thesis: Steric exclusion effects and biochemical activity of molecular motors Adéla Brucknerová, 2022 The cytoskeleton and its associated proteins, the so-called molecular motors, belong to the cellular apparatus of every eukaryotic cell. Molecular motors, including kinesin-1, have the ability to temporarily bind to the cytoskeleton and transport cargo vesicles (e.g. organelles) along it. During such movement, kinesin-1 molecules hydrolyse one ATP molecule for each step of their chemomechanical cycle. It turns out, that the chemomechanical cycle may be affected by the presence of inert macromolecules. The influence of these macromolecules on the chemomechanical cycle of kinesin-1 is, however, not entirely known. Using total internal reflection fluorescence microscopy, single molecules of kinesin-1 were tracked in vitro. Their movement along reconstituted microtubules was recorded and the average velocity was measured. The performed experiments were carried out with variety of concentrations of soluted macromolecules - polyethylene glycol. For experiments, polyethylene glycols of molecular weight 6 kg/mol, 1000 kg/mol and combination of both were used. Characterization of newly developed fluorescence probes in cellular systems

Nanoparticles (NP) are currently a progressive area of scientific research. The possibility of synthesizing them according to the required parameters opens up possibilities for their wide use also in biomedicine. One example is a nanoparticle that can detect cellular processes, such as pH. We already know that the pH of healthy and cancer cells differs by the opposite gradient on the intracellular and extracellular side of the membrane. In this context, this work deals with the study of fluorescent silicon nanoparticles (SiNP) tested on a human keratinocyte cell line from a healthy donor (HaCaT) and from skin cancer donor (A431). Once found that even the highest concentrations of SiNP used are not cytotoxic, they can be further studied by fluorescence, confocal and super-resolution microscopy. In order to assess the pH detection properties of these SiNPs, a method for measuring intracellular pH with a fluorescent raciometric probe SNARF-1 using fluorescence spectroscopy and flow cytometry was introduced. Theoretical part is focused on ERP, CRM, SCM systems and application software. Analytical part deals with investments of czech SME in software and ICT equipment. Interakce xenobiotik s MDR pumpami u S. cerevisiae

The interactions of a number of xenobiotics with two main S. cerevisiae MDR pumps, Pdr5p and Snq2p were studied. The results showed that some compounds do not interact with MDR pumps whereas others affect one or another of the pumps, causing a loss of their ability to transport an indicator substrate out of the cells. This apparent inactivation of the pumps can be due to competition for transport with the indicator substrate or to actual (noncompetitive) inhibition of the pumps
