Dopad alimentární expozice bisfenolu na vybrané krevní parametry samic potkanů kmene Wistar Han

This bachelor's thesis focused on assessing the impact of ten-week oral exposure to 50 ?g BPS/kg body weight per day in female Wistar Han rats on the concentration of hemoglobin (HGB), hematocrit value (HCT), mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), mean corpuscular hemoglobin (MCH), red blood cell count (RBC), white blood cell count (WBC), percentage of granulocytes (GR), lymphocytes (LY), and monocytes (MONO).

Bisphenol S (BPS) is an organic substance which has replaced significantly industrially used bisfenol A (BPA). Eventhough BPS shows higher tolerance against physical influence, there have been registered leaks from the final products and there is a posibility of wather, food and environtment contamination. Until now the BPS has been, just like BPA, mostly connected with disruption of the reproductive system of human and animals. The aim of the bachelor thesis was to determinate whether a ten weeks oral exposure to BPS (50 ?g, 100 mg/kg live weight/day) leads to disturbance in selected haematological parameters (erythrocyte count (RBC), haemoglobin concentration (HGB), haematocrit value (HCT), mean corpuscular volume (MCV), erythrocyte haemoglobin concentration (MCHC), mean corpuscular haemoglobin (MCH), total leucocytes, percentage of granulocytes, lymphocytes and monocytes). Eighteen Wistar Han rats were enrolled in the experiment and they were divided into three groups (control group, 50 ?g BPS, 100 mg BPS). Dissolved BPS in sunflower oil was given to the experimental groupts by a gavage. Blood was collected through cardiac puncture and analyzed using an automated hematology analyser. In the particular study, we found out a nonsignificant decrease of HGB and nonsignificant increase of MCV and MCH in experimental animals. BPS also led to a nonsignificant changes in the number of leukocytes (increase in the total number of leukocytes) and it led to a changes in the number of individual types of the white blood cells representation (increase in the percentage of granulocytes, decrease in lymphocytes). More significant effect was observed in 50 ?g BPS group. Our results did not conclusively proved the negative effect of BPS on the selected blood parameters of rats.
Vliv supranutričních dávek organické formy selenu na vybrané krevní parametry králíků

Selenium is an important essential mikroelement, that acts through a number of selenoproteins. Both it´s lack, and it´s plenty has a negative influence on the organism. Recently is used Se during the various diseases treatment of people with supranutritive dosages of this element. The presented thesis deals with the supranutritive dosages influence of Selenium concentration organic form (Chlorella, enlarged alga with selenium) in blood serum and selected rabbits blood parameters (hemoglobin and hematocrit level, leukocytes and erythrocytes, urea concentration, triglycerides, gamma value glutamyltransfers and total concentration of plasmatical proteins). Nine week experiment was performed on 18 rabbits, which were divided in to three experimental groups. Group 1 (control group) has received the selenium at a dosage of 0.15 mg / kg dry matter of feed, group 2 at a dosage of 0.3 mg / kg dry matter feed and group 3 at a dosage of 4.3 mg / kg of dry matter feed. During the experiment weren´t registered any clinical symptoms associated with the possible Se toxicity. Supranutritive dosages in the group 3 led to faster and statistically significant increase of the selenium concentration in the blood serum. Parameters of the hemopoiesis, the growth and the substantial metabolism had no influnece with supranutritive dosages in the groups 2 and 3. The results has not confirmed the clear positive or negative effect of selenium supranutritive dosages on selected blood parameters. The results show, that after rabbits supplementation are suitable recommended dosages of 0.15 mg Se / kg dry matter feed.
Dynamics of selected hematological and biochemical parameters in blood plasma of calves in relation to environmental conditions.

The aim of my thesis is to evaluated the dynamics of selected hematological and biochemical parameters in blood plasma of calves reared in different environmental conditions. Totally were collected 120 blood samples of calves in age of 1 to 124 days. The animals came from three different breeds with different ways of rearing. The total blood parameters tested in all observed groups were in the normal range. Dynamics of selected hematological and biochemical parameters in blood plasma of calves in all groups were relatively similar. It is clear that the calves, while preserving the basic animal hygiene conditions of livestock, can cope with the effects of different technologies of rearing and environmental conditions.
