Agrotechnics cultivation and content of some biologically active substances in plants of the genus Echinacea

This thesis describes the effect of agrotechnics on biologically active substances in Echinacea species. It deals with botanical characteristic, cultivation, agrotechnics and protection against pests. In the second part are described the chemical composition and active substances of Echinacea species and their options to improves the substances in the plants.

Blastocystis in domestic birds

During 2010, faeces samples for parasitology research oriented on Blastocystis were collected from an anonymous farm. A total of 55 samples were collected (of them for 10 faeces samples from hens, ducks and geese and 25 from pigeons). The samples were cultured in Dobell-Leidlaw medium and viewed by light microscopy. However, this method led to only two foundings of Blastocystis (one from hen, the other from pigeon). Thus, PCR and nested-PCR with specific primers were later used for better detection of Blastocystis. The samples for PCR detection were not collected from the aforementioned farm, but they originated from a collection of isolated DNA samples that was available on the Parasitology institute of AS CR. The results of PCR were checked after electrophoresis and verified by sequenation. The obtained sequences of bird-isolated Blastocystis were phylogenetically analysed and as described to subtype 7.

This M.Sc. thesis is aimed to describe growth and production parameters of nematophagous fungus Arthrobotrys oligospora. There was determinated information about growth and sporulation of fungus on artificial and natural substrates, influence of cultivation condition on production biomass and possibility of formulation alginate pellets.
Biofuels from algae: Physiological characterization of candidate diatom species

The aim of this study was to isolate and extensively test promising candidate diatom species for biodiesel cultivation. Diatom strains were isolated from natural habitats in the state of Ohio (USA). These strains were tested to find optimal growing conditions and media (concentration of Nitrogen, Phosphorus, Silica, source of Nitrogen, temperature etc.).
Profile measurements of soil processed.

The aim of the bachelor's thesis is to create an overview of soil profile measuring ways and as a result of its analysis to design and construct a measuring device which would enable to record measured values for further computer processing. Cultivation is one of the most energy-demanding procedures in the crop production. It is possible to choose implements with various energy demand, but at the same way it is necessary to comply with the required agricultural conditions for a particular type of crop. To meet both requirements, it is desirable to ascertain the actual effect of the tool in soil or a real processed profile. The proposed device can detect the depth of cultivation, soil profile and soil looseness. These values serve to determine the working quality of complement, its setting and furthermore they are used for calculating the economic indicators of soil cultivation.
In vitro cultivation possibilities of orchid Macodes petola

The orchid family covers 90 percent of the rarest plants under protection all over the world. The ecological and economic value of orchids has aroused public awareness. Since vegatitve propagation methods are available, development of in vitro methods are essential for conservation and commercialization of plants. Purposes work was elaboration progress of vegetative multiplication for tropical orchid Macodes petola in conditions in vitro. At research were proof 4 methods, which distinguish in using desinfection, in operating time and in using different nutritive mediums.
Kultivace mikrořas v trubicovém fotobioreaktoru založeném na solárních koncentrátorech: distribuce světla

The cultivation system consists of two types of irradiance modules – vertical, lower-irradiance unit (close to ambient conditions) and roof, higher-irradiance unit (up to 3.5-times higher than ambient intensity) which are used to compare the growth and behaviour of microalgal cultures of various density at high light intensities. The aim of these experiments has been to monitor a diel course of irradiance intensity and spectral composition concentrated by Linear Fresnel lenses and the light distribution on the surface and inside cultivation tubes. As a model organism, we have used the cyanobacterium Spirulina platensis M2. Three concentration of biomass (about 0.5, 1 and 2 g L-1) have been tested. In higher-irradiance modules we also follow the induction of carotenoids synthesis
