A novel structurally characterized haloacid dehalogenase superfamily phosphatase from Thermococcus thioreducens with diverse substrate specificity

This RNDr. thesis is focused on complex studies of a haloacid dehalogenase superfamily phosphatase member with an unknown substrate specificity, isolated from a hyperthermofilic archeon Thermococcus thioreducens. The HAD phosphatase, internally named as Tt82, was isolated, expressed and purified using standard techniques and the crystal structure was solved by X-ray crystallography method. The refined crystal structure displayed characteristic HAD superfamily elements, such as the HAD canonical fold with the HAD signature motifs in the active site. Further, possible substrates for the enzyme were proposed and examined by computational docking in the active site of the HAD phosphatase. The results of the docking studies served as a basis for an experimental measurement of Tt82 enzymatic activity against selected substrates at two different temperatures. HAD phosphatase Tt82 is a promising target for the biotechnological field, mainly for its wide substrate specificity and higher temperature optima.
The use of Musical Instrument Djembe in Work with Clients of Educational Child Care Center

This bacelor thesis deals with the use of the musical instrument called the djembe by the clients of the Centre for Educational Care. Two goals were determined in this work. The first goal was to "Develop a proposal for an educational program of leisure activities using the djembe for the clients of the Centre for Educational Care" and the second goal was to "Map the possibilities of using the djembe in creating the leisure programs for the clients of the Centre of Educational Care". The thesis consists of the theoretical and practical part. The theoretical part includes four chapters. In addition to the characteristics of special education of children with behavioural disorders and its position in special pedagogy, the first chapter specifies behavioural disorders and the factors that influence their origin, and finally it describes the Centre for Educational Care. The second part is devoted to the characteristics of leisure activities in general and their function in influencing the clients with behavioural disorders. The topic of the third chapter is music therapy and its use in clients with behavioural disorders, including drumming. The fourth chapter introduces the djembe and the principles of drumming. The practical part describes the research methodology, actual implementation and the results of the research. The methods of qualitative research such as observation, interviews and secondary analysis of the documents were used as the research methods. The author prepared a 180 minute program for pilot verification. Based on this verification, a 60 minute program was compiled and tested with three different groups of clients. Following implementation of the 60 minute programs, it was found that the time allocation for these programs was insufficient. Therefore, the final 90 minute version was prepared and tested. A part of the thesis is the annex that contains suggestions for the relaxation parts of the djembe drumming program. In addition to the final 90 minute version of the program, the chapter with the results also includes conclusions from interviews with the employees of the Centre for Educational Care regarding further use of the djembe.

The thesis analyses the Institute of the President of the Czech Republic in the Czech and Slovak Republics from 1993 to 2017. Main focus of the thesis is the comparison of the position of the Presidents of the Czech Republic and the Slovak Republic. The thesis look closer general position of head of states in the political systems, the election of the president, the introduction of a direct election in the Czech Republic, their authority, responsibility, termination of service and real application of presidential powers. The diploma thesis focuses on the media image of the presidents, their popularity, media cases and public opinion. The thesis also aims at the organized comparison of the powers and position of the presidents of both countries between 1993 and 2017, identification of identical or different features. Effects of plants on the structure, function and diversity of bacterial communities

Vegetation is known to influence the composition of microbial communities. Bacteria can act as roots symbionts or be involved in the decomposition of plant biomass. They can be influenced by soil chemistry but also by plant exudates. Some plants produce targeted exudates to attract specific bacteria to their roots. Bacteria associate with plants frequently but the effect of plant diversity on bacterial communities on their roots and in the surrounding soil remains unclear. The aim of this work was to describe the relationship between the diversity and community composition of bacteria and the diversity of vegetation in forest and grassland ecosystems. The study areas were selected to represent a gradient of vegetation in Bohemian Forest NP and in White Carpathian flowery grasslands. I hypothesized that the diversity and evenness of bacterial community increase with increasing plant diversity. The composition of bacterial community was characterized by 16S rRNA sequencing. The composition of vegetation was determined by phytocenological relevées and by molecular markers trnL. In grassland ecosystem, there was a positive relationship between plant and bacterial diversity only in shoots. Dissemination of bacterial resistance to tetracycline antibiotics from animal husbandry to the soil.

This bachaleor thesis is dedicated to occurence of tetracycline resistance genes in the soil environment influenced by wastes from a livestock productionanimal husbandry. In the experimental part, the transfer of tetracycline resistance genes from the excrements of a dairy cattle to the soils was studied.
