[cze][eng] Nové psychoaktivní substance (NPS) představují skupinu heterogenních látek s širokým spektrem farmakologických účinků - mohou působit stimulačně, tlumivě, vyvolávat pocity emoční blízkosti (tzv. entaktogenní účinek), působit halucinogenně nebo jinak měnit vnímání reality. Označení "nové" se přitom nutně nemusí vztahovat k jejich chemické struktuře, ta může být známa již řadu let, ale spíše k jejich relativně nedávnému výskytu na ilegálním trhu. U většiny NPS zůstávají jejich farmakokinetické vlastnosti, toxicita, behaviorální účinky i adiktivní potenciál nedostatečně prozkoumány, což představuje významné riziko pro veřejné zdraví. Cílem dizertační práce bylo experimentální studium vybraných NPS v animálním modelu. Jednotlivé cíle byly následující: 1) Stanovení farmakokinetiky, metabolomiky a toxicity vybraných NPS reprezentovaných disociativními anestetiky deschlorketaminem a methoxfenidinem, syntetickým katinonem nafyronem, fenethylaminovým psychedelikem 25CN-NBOMe a tryptaminovým psychedelikem baeocystinem; 2) Charakterizace jejich behaviorálních účinků a 3) Evaluace adiktivního potenciálu nafyronu. Látky byly subkutánně aplikovány potkanům kmene Wistar v rozmezí dávek odvozeném z dostupných literárních údajů a, pokud byly k dispozici, také z přepočtu dávek užívaných u lidí, v následujících...New psychoactive substances (NPS) represent a group of heterogeneous substances with a wide range of pharmacological effects - they can have stimulating or sedative effects, induce feelings of emotional closeness (the so-called entactogenic effect), cause hallucinations, or otherwise alter the perception of reality. The term "new" does not necessarily refer to their chemical structure, which may have been known for many years, but rather to their relatively recent appearance on the illegal market. For most NPS, their pharmacokinetic properties, toxicity, behavioral effects, and addictive potential remain insufficiently studied, posing a significant risk to public health. The aim of the dissertation was to experimentally study selected NPS in an animal model. The individual objectives were as follows: 1) Determination of the pharmacokinetics, metabolomics, and toxicity of selected NPS represented by the dissociative anesthetics deschloroketamine and methoxphenidine, the synthetic cathinone naphyrone, the phenethylamine psychedelic 25CN-NBOMe, and the tryptamine psychedelic baeocystin; 2) Characterization of their behavioral effects; and 3) Evaluation of the addictive potential of naphyrone.
