Tomáš Havrišák Univerzita Karlova, Farmaceutická fakulta v Hradci Králové Skin Barrier Research Group, Katedra organické chemie a bioorganické chemie Stratum corneum (SC) představuje klíčovou část kožní bariéry, nacházející se na úplném povrchu epidermis, která podléhá neustálému procesu obnovy. SC je tvořeno zrohovatělými buňkami (korneocyty), lipidy a desmozomy, přičemž lipidová matrix je klíčová pro udržení bariérové funkce. Hlavními složkami těchto lipidů jsou ceramidy, cholesterol a volné mastné kyseliny. Ceramidy hrají zásadní roli v ochraně kůže a jejich základní složkou jsou sfingoidní báze, alifatické aminoalkoholy s dlouhým řetězcem, které tvoří spolu s mastnou kyselinou základní stavební bloky sfingolipidů. V kožní bariéře se nacházejí čtyři základní sfingoidní báze: sfingosin, dihydrosfingosin, fytosfingosin a 6-hydroxysfingosin. Tyto sloučeniny se liší přítomností dvojných vazeb a hydroxylových skupin, přičemž každá z nich přispívá k funkci a struktuře ceramidů v kůži. Tomáš Havrišák Charles University, Faculty of Pharmacy in Hradec Králové Skin Barrier Research Group, Department of Organic and Bioorganic Chemistry The stratum corneum (SC) represents a key part of the skin barrier, located on the entire surface of the epidermis, which is subject to a constant renewal process. The SC is made up of corneocytes, lipids and desmosomes, with the lipid matrix being crucial for maintaining barrier function. The main components of these lipids are ceramides, cholesterol and free fatty acids. Ceramides play a vital role in skin protection and their basic components are sphingoid bases, aliphatic amino alcohols with a long chain, which together with fatty acids form the basic building blocks of sphingolipids. There are four basic sphingoid bases in the skin barrier: sphingosine, dihydrosphingosine, phytosphingosine and 6-hydroxysphingosine. These compounds differ in the presence of double bonds and hydroxyl groups, each contributing to the function and structure of ceramides in the skin.
