[cze][eng] Biostimulanty jsou látky s širokou možností využití v moderním zemědělství pro podporu růstu a odolnosti plodin. Tato bakalářská práce se zabývá studiem transportu a metabolického osudu biostimulantu MTU radioaktivně značeného tritiem (3 H-MTU), v rostlinách pšenice seté (Triticum aestivum L., odrůda Balitus). Cílem práce bylo sledování dynamiky akumulace této značené látky v rostlinných pletivech a identifikace jejích případných metabolických produktů pomocí kombinace metod autoradiografie a vysokoúčinné kapalinové chromatografie s radiometrickou detekcí (radio-HPLC). Pro prvotní ověření distribuce biostimulantu byla využita metoda autoradiografie. Pro následnou chromatografickou analýzu byly optimalizovány podmínky separace a přípravy vzorků. K purifikaci a zakoncentrování rostlinných extraktů byla zvolena extrakce na pevné fázi (SPE) zajišťující maximální výtěžek analytu v první methanolové frakci. Chromatografická separace probíhala v izokratickém módu na analytické koloně XBridgeTM C18 (4,6 x 150 mm, 3,5µm) při průtokové rychlosti mobilní fáze 0,5 ml/min. Mobilní fáze byla složena ze 75 % (v/v) vodné složky A (0,1% (v/v) vodný roztok kyseliny mravenčí) a 25 % (v/v) organické složky B (methanol). Zvolený poměr zajistil optimální retenci mateřské látky s retenčním časem přibližně 10 minut a...Biostimulants are substances with a wide range of applications in modern agriculture to promote crop growth and resistance. This bachelor thesis deals with the study of the transport and metabolic fate of the biostimulant MTU, radiolabelled with tritium (3 H-MTU), in wheat plants (Triticum aestivum L., var. Balitus). The aim of the work was to monitor the dynamics of accumulation of this labelled substance in plant tissues and to identify its possible metabolic products using a combination of autoradiography and high-performance liquid chromatography with radiometric detection (radio-HPLC). Autoradiography was used for the initial verification of the biostimulant distribution. For the subsequent chromatographic analysis, the conditions of separation and sample preparation were optimized. Solid-phase extraction (SPE) was chosen for the purification and concentration of plant extracts, ensuring the maximum yield of the analyte in the first methanol fraction. Chromatographic separation was performed in an isocratic mode on an XBridge™ C18 analytical column (4.6 × 150 mm, 3.5 µm) at a mobile phase flow rate of 0.5 ml/min.
