[cze][eng] Tato bakalářská práce se zabývá studiem derivatizačních činidel pro analýzu méně polárních lipidů obsahujících volnou hydroxylovou nebo karboxylovou skupinu analyzovaných pomocí hydrofilní interakční chromatografie ve spojení s hmotnostní spektrometrií. Teoretická část práce se zaměřuje na obecnou charakteristiku lipidů, přehled analytických metod používaných v lipidomice a derivatizační reakce vybraných tříd lipidů. Cílem experimentální části bylo navrhnout derivatizační činidla vhodná pro derivatizaci vybraných méně polárních lipidových tříd a zhodnotit přínos derivatizace pro jejich stanovení. Hodnocení bylo provedeno na základě vlivu na vybrané standardy glycerolipidů, mastných kyselin a volných sterolů, se zaměřením na retenční chování v režimu HILIC, ionizaci a charakteristické strukturní fragmenty vznikající pomocí tandemové hmotnostní spektrometrie. V závěrečné části jsou shrnuty výhody derivatizace a potenciál jednotlivých činidel pro analýzu vybraných lipidových tříd. Klíčová slova: chemická derivatizace, lipidy, hydrofilní interakční chromatografie, hmotnostní spektrometrie, glycerolipidy, steroly, mastné kyselinyThis bachelor's thesis deals with the study of derivatization reagents for the analysis of less polar lipids containing a free hydroxyl or carboxyl group analyzed using hydrophilic interaction chromatography coupled with mass spectrometry. The theoretical part of the work focuses on the general characteristics of lipids, an overview of analytical methods used in lipidomics and derivatization reactions of selected lipid classes. The aim of the experimental part was to design derivatization reagents suitable for the derivatization of selected less polar lipid classes and to evaluate the benefit of derivatization for their determination. The evaluation was performed based on the effect of selected standards of glycerolipids, fatty acids and free sterols, focusing on retention behavior in the HILIC mode, ionization and characteristic structural fragments generated using tandem mass spectrometry. The final part summarizes the advantages of derivatization and the potential of individual reagents for the analysis of selected lipid classes.
