PharmDr. Zdeňka Šklubalová, Ph.D. Consultant: Sebastien Bailey Student: Michaela Stanková Title of Thesis: Evaluation of the influence of binary lubricant mixtures on the compression of model tableting blend Lubricants are essential excipients in tablet formulations, as they reduce friction between the tablet and the die during ejection, thereby preventing their mechanical damage. However, lubricants may adversely affect the mechanical properties of tablets. This study evaluates the impact of using two lubricants, magnesium stearate (MgSt) and sodium stearyl fumarate (SSF) in binary combinations, on tablet ejection force and tablet properties. Powder blends consisting of microcrystalline cellulose and lactose (MCC/L 1:1) were used as a model filler. MgSt was incorporated at base concentrations of 0,5% or 0.25%, and SSF was added at MgSt:SSF ratios 1:1, 1:2, 1:3 and 1:4. All mixtures exhibited poor flow propertie according to the Hausner ratio, with values ranging from 1.46 to 1.50. The blends were compressed into tablets at 10 kN and ejection force was measured. Znižujú trenie medzi tabletou a matricou počas jej vytláčania, čím zabraňujú ich poškodeniu. Nanešťastie však lubrikanty môžu negatívne ovplyvňovať mechanické vlastnosti tabliet. Táto štúdia hodnotí vplyv binárnych zmesí lubrikantov stearanu horečnatého (MgSt) a stearyl fumarátu sodného (SSF) na sypné a lisovacie charakteristiky modelovej tabletoviny a vlastnosti tablet. Tabletovina bola pripravená z mikrokryštalickej celulózy a laktózy (MCC/L) v pomere 1:1 s prídavkom MgSt 0,5% alebo 0.25% a SSF tak, aby bol dosiahnutý pomer 1:1, 1:2, 1:3 a 1:4. Podľa hodnoty Hausnerovho pomeru v rozmedzí 1,46-1,50 mali binárne zmesi veľmi zlé tokové vlastnosti. Tablety boli lisované silou 10 kN a bola meraná sila potrebná na ich vytlačenie. Po 24 hodinách sa hodnotila pevnosť v ťahu, čas rozpadu a oder tabliet.
