[cze][eng] Patogeneze metabolického syndromu (MetS) zahrnuje složité interakce mezi genotypem, epigenetikou a faktory životního stylu. Tato diplomová práce zkoumá, jak rozvoj MetS ovlivňují vysokotukové diety s odlišným obsahem sacharidů. Konkrétně se věnuje vlivu vysokotukových diet na morfometrické a metabolické parametry u experimentálního modelu metabolického syndromu - inbredního spontánně hypertenzního kmene potkana SHR. Cílem této práce je objasnit, zda konzumace stravy s vysokým obsahem tuku a sacharózy ovlivňuje metabolické parametry a distribuci tukové tkáně u obou pohlaví. Dalším cílem je zhodnotit, jaké jsou rozdíly v odpovědi organismu na tyto dietní intervence v závislosti na pohlaví. Do výzkumu bylo zahrnuto 36 dospělých potkanů kmene SHR rozdělených do šesti skupin podle pohlaví a typu podávané diety: standardní (STD), vysokotukové bez sacharózy (HFD) a vysokotukové se sacharózou (HFDS). Všechny skupiny byly po dobu 4 týdnů sledovány a následně u nich byly provedeny morfometrické a laboratorní analýzy. Statisticky významný efekt typu diety byl potvrzen u hmotnosti dep tukové tkáně, hladin sérového leptinu a lipidů v séru a jaterní tkáni. Mezi pohlavími byl prokázán vysoce signifikantní rozdíl v celkové tělesné hmotnosti i v relativní hmotnosti vnitřních orgánů, jako jsou srdce, játra a...The pathogenesis of metabolic syndrome (MetS) involves complex interactions between genotype, epigenetics, and lifestyle factors. This master's thesis examines how the development of MetS is influenced by high-fat diets with varying carbohydrate content. Specifically, it focuses on the effects of high-fat diets on morphometric and metabolic parameters in an experimental model of metabolic syndrome - the inbred spontaneously hypertensive rat strain SHR. The aim of this study is to clarify whether the consumption of a diet high in fat and sucrose affects metabolic parameters and adipose tissue distribution in both sexes. A further objective is to evaluate the sex-dependent differences in the organism's response to these dietary interventions. The study included 36 SHR rats divided into six groups based on sex and diet type: standard (STD), high-fat without sucrose (HFD), and high-fat with sucrose (HFDS). All groups were monitored for 4 weeks, followed by morphometric and laboratory analyses. A statistically significant diet-related effect was confirmed for adipose tissue mass, serum leptin levels, and lipid levels in serum and liver tissue.
