Účinek agonistů nukleárních receptorů (FXR, CAR, aj.) na játra a hepatocyty in vivo a in vitro za různých stavů Metabolickou dysfunkcí podmíněné steatotické onemocnění jater (MASLD) postihuje významnou část světové populace, přičemž v současné době jsou farmakoterapeutické možnosti omezené. Mitochondriální dysfunkce představuje jeden z klíčových patofyziologických mechanizmů rozvoje a progrese tohoto onemocnění. Farnesoid X receptor (FXR) je významným regulátorem metabolizmu lipidů, sacharidů i žlučových kyselin a jeho aktivace může ovlivňovat metabolické a buněčné děje relevantní pro MASLD. Agonisté FXR jsou zkoumáni jako potenciální terapeutická možnost u MASLD, avšak jejich přímé účinky na jaterní mitochondriálníbioenergetikunejsoudosud plně objasněny. Cílem této disertační práce bylo komplexně zhodnotit vliv FXR agonisty, kyseliny obeticholové (OCA), na mitochondriální respiraci v podmínkách MASLD pomocí in vivo i in vitro modelů. K in vivo experimentu byly použity kontrolní myši a myši s MASLD indukovaným 36týdenním podáváním diety západního typu obohacené o glukózu a fruktózu v pitné vodě. V posledních třech týdnech byla zvířatům podávána OCA v dávce 5 nebo 10 mg/kg/den. Na konci experimentu byla hodnocena jaterní histologie, biochemické parametry, genová exprese, aktivita mitochondriálních enzymů a... The farnesoid X receptor (FXR) is an important regulator of lipid, carbohydrate,and bile acid metabolism,and its activationmayinfluence metabolic andcellular processesrelevant toMASLD.FXR agonists are therefore being investigated as a potential therapeutic option for MASLD; however, their direct effects on hepatic mitochondrial bioenergetics remainincompletelyunderstood.The aim of this dissertation was to evaluate the effect of the FXR agonist obeticholic acid (OCA) on mitochondrial respirationunder MASLD conditions using both in vivo and in vitro models.
