Genová terapie představuje moderní medicínský přístup využívající metody genového inženýrství k léčbě dědičných i získaných onemocnění na molekulární úrovni. Cílem této bakalářské práce je zmapování rychlého vývoje tohoto oboru, ke kterému došlo v několika posledních dekádách, a především zaměření na aktuálně využívané metody. Postup genové terapie probíhal od využití tradičních virových vektorů k vývoji bezpečnějších a efektivnějších nanočástic využívaných pro precizní editaci genomu pomocí metod, jako je prime editing nebo base editing. V současné době existují schválené genové terapie pro široké spektrum vzácných onemocnění postihující různé části těla. Aktuální výzkumy probíhající na poli genové terapie jsou zaměřeny především na rozšíření využitelnosti již existujících metod pro onemocnění postihující větší množství lidí a také na zapojení nových technologií, jako je umělá inteligence, do vývoje. klíčová slova: genová terapie, editace genomu, virové vektory, lipidové nanočástice, CRISPR/Cas, dědičná onemocnění, léčba Gene therapy represents a modern medical approach utilizing genetic ingeneering methods to treat hereditary and acquired diseases at the moleclar level. The aim of this bachelor's thesis is to map the rapid evolution of this field over the past several decades, with a primary focus on currently used methods. The field of gene therapy has evolved from the use od traditional viral vectors toward the creation of safer and more efficient nanoparticles used for precise genome editing via methods such as prime editing or base editing. Currently, approved gene therapies exist for a wide spectrum of rare diseases affecting various parts of the human body. Ongoing researchis focused mainly on expanding the applicability of existing methods to diseases affecting a larger population and on the intergation of new technologies, such as artificial inteligence, into development. key words: gene therapy, genome editing, viral vectors, lipid nanoparticles, CRISPR/Cas, hereditary diseases, treatment
