[cze][eng] Cirkadiánní rytmicitu vykazuje řada fyziologických procesů, což potvrzuje i rozmanité spektrum zdravotních komplikací vznikajících v důsledku jejich desynchronizace s rytmy vnějšího prostředí. Tato práce se zaměřuje na cirkadiánní systém člověka a poruchy postihující zejména cyklus spánku a bdění. Práce nejprve charakterizuje molekulární mechanismy generování biologických rytmů a roli suprachiasmatického jádra. Následně hodnotí terapeutické využití exogenního melatoninu, který plní roli substituce deficientní či desynchronizované sekrece endogenního hormonu. V neposlední řadě je zdůrazněn význam správného načasování administrace, které je klíčové pro efektivní resynchronizaci vnitřních hodin u pacientů s cirkadiánními poruchami. This work focuses on the human circadian system and disorders that are associated specifically with the sleep-wake cycle. The work first characterizes the molecular mechanism of biological rhythm generation and the role of the suprachiasmatic nucleus. Secondly it evaluates the terapeutic use of exogenous melatonin, which serves as a substitution for deficient or desynchronized endogenous hormone. Finally, the importance of correct timing administration is emphasized, as it is crucial for the effective resynchronization of the internal clock in patients with circadian disorders.
