[cze][eng] Hemato-testikulární bariéra dělí semenotvorný epitel na bazální a apikální kompartment, přičemž v apikálním kompartmentu se nachází imunologicky privilegované mikroprostředí nezbytné pro dokončení diferenciace zárodečných buněk. Bariéra je tvořena těsnými spoji sousedících Sertoliho buněk, které vznikají díky integrálním membránovým proteinům jako jsou klaudiny, okludin a JAMs. Konkrétně klaudin-11 se ukázal být esenciální pro tvorbu vláken těsných spojů Sertoliho buněk. Klíčové proteiny těsných spojů jsou pomocí scaffold proteinů fyzicky napojené na buněčný cytoskelet, který se účastní dynamické přestavby těsných spojů. Se spoji asociovaná aktinová filamenta jsou uspořádána do stabilních svazků, které zajišťují integritu bariéry. Během cyklické remodelace hemato-testikulární bariéry se aktinové svazky mění na dynamickou větvenou síť, která umožňuje internalizaci integrálních membránových proteinů spojů. Přeměna struktury aktinových filament je zprostředkována řadou aktin-vazebných proteinů. Intermediální filamenta činí Sertoliho buňky odolnější vůči mechanickému stresu a deformacím a mají vliv na stabilitu spojovacího komplexu. V průběhu semenotvorného epiteliálního cyklu dochází k transportu preleptotenních spermatocytů přes hemato-testikulární bariéru, aniž by docházelo k průniku látek zvenčí do...The blood-testis barrier divides the seminiferous epithelium into basal and apical compartments, with the apical compartment forming an immunologically privileged microenvironment essential for the completion of germ cell differentiation. The barrier is formed by tight junctions between adjacent Sertoli cells, which arise from integral membrane proteins such as claudins, occludin, and junctional adhesion molecules (JAMs). In particular, claudin-11 has been shown to be essential for the formation of Sertoli cell tight junction strands. Key tight junction proteins are physically linked to the cellular cytoskeleton via scaffold proteins, which participate in the dynamic remodelling of tight junctions. Actin filaments associated with these junctions are organized into stable bundles that ensure barrier integrity. During the cyclic remodelling of the blood-testis barrier, these actin bundles are reorganized into a dynamic branched network, enabling the internalization of integral membrane junctional proteins. This transformation of actin filament structures is mediated by a range of actin- binding proteins. Intermediate filaments render Sertoli cells more resistant to mechanical stress and deformation and contribute to the stability of the junctional complex. During the seminiferous epithelial cycle,...
