[cze][eng] Spermie představuje vysoce specializovanou buňku, která je pokryta hustou sacharidovou vrstvou nazývanou glykokalyx, kterou tvoří glykanové řetězce navázané na proteiny a lipidy spermie. Významným monosacharidem v těchto řetězcích je sialová kyselina, která se nejčastěji vyskytuje na terminální pozici. Kyselina sialová má v procesu oplození mnoho klíčových rolí, od komunikace spermie s imunitním systémem přes její transport reprodukčním traktem a regulaci načasování kapacitace až po interakci gamet při oplození. Aby však tyto procesy mohly být zprostředkovány, jsou zapotřebí specifické proteiny. Jedná se především o enzymy neuraminidázy a sialyltransferázy podílející se na dynamické regulaci sialylace glykanů. Kromě enzymatické regulace jsou důležité také proteiny schopné rozpoznávat sialylované struktury. Tyto proteiny jsou označované jako lektiny a patří mezi ně například lektin vázající kyselinu sialovou se strukturním motivem podobným imunoglobulinu a selektiny, které po rozpoznání sialové kyseliny ovlivňují molekulární dráhy potřebné pro úspěšné oplodnění. Cílem této prácebyla charakterizace enzymů a lektinů spojených se sialylací. Pochopení těchto mechanismů nám umožní objasnit průběh klíčových kroků reprodukčního procesu. Klíčová slova Kyselina sialová, sialyltransferázy, neuraminidázy,...The sperm is a highly specialized cell covered by a dense carbohydrate layer called the glycocalyx, which consists of saccharides bound to sperm proteins and lipids. A significant monosaccharide within these chains is sialic acid, which is most frequently located at the terminal position. Sialic acid plays many key roles in the fertilization process, ranging from the communication of the sperm with the immune system, through its transport within the reproductive tract and the regulation of capacitation, to the interaction of gametes during fertilization. However, these processes are mediated by specific proteins, primarily the enzymes neuraminidases and sialyltransferases, which jointly regulate the modification of glycan sialylation. Beyond its enzymatic regulation on carbohydrate chains, proteins that recognize sialic acid play a crucial role. These proteins are referred to as lectins, and they include, for example, sialic acid-binding immunoglobulin-like lectins and selectins, which recognize sialylated structures and influence the subsequent molecular pathways necessary for successful fertilization. The aim of this thesis was the characterization of these enzymes and lectins, the understanding of which allows us to elucidate the course of key steps in the reproductive process. Keywords Sialic...
