[cze][eng] Tato bakalářská práce shrnuje dostupnou literaturu o agonistech receptoru pro glukagonu podobný peptid- 1. Práce popisuje strukturu a fyziologii glukagonu podobného peptidu-1, mechanismus aktivace jeho receptoru a farmakologické vlastnosti jednotlivých agonistů. Pozornost je věnována strukturním modifikacím prodlužujícím biologický poločas, které umožnily vývoj látek od krátkodobě působících analogů až po moderní dlouhodobě působící multiagonisty. Vedle využití v léčbě diabetu mellitu 2. typu a obezity jsou diskutovány pleiotropní účinky zahrnující kardiovaskulární protekci, neuroprotektivní potenciál v kontextu Alzheimerovy a Parkinsonovy choroby či hepatoprotektivní působení. Práce mapuje také aktuální směry výzkumu včetně perorálních nízkomolekulárních agonistů. The thesis describes the structure and physiology of glucagon-like peptide-1, the receptor activation mechanism and pharmacological properties of individual agonists. Structural modifications extending biological half- life are discussed, enabling development from short-acting analogues to modern long-acting multi-agonists. Beyond type 2 diabetes and obesity treatment, pleiotropic effects are reviewed including cardiovascular protection, neuroprotective potential in Alzheimer's and Parkinson's disease, and hepatoprotective effects.
