[cze][eng] Kingella kingae je pediatricky významný bakteriální patogen, jehož klíčovým faktorem virulence je cytotoxin RtxA z rodiny RTX toxinů. Pro přechod neaktivního protoxinu do biologicky aktivní formy proteinu je nezbytná posttranslační modifikace specifickou acyltransferasou RtxC a následné navázání vápenatých iontů do RTX domény. Cílem této bakalářské práce bylo prozkoumat nedávno predikovaná vazebná místa pro vápenaté ionty lokalizovaná v acylovaném segmentu toxinu RtxA a rozšířit tak poznatky o mechanismu jeho skládání do aktivní konformace v extracelulárním prostředí. Pomocí cílené mutageneze bylo připraveno 15 mutantních variant toxinu RtxA s bodovými mutacemi v oblastech předpokládaných β-otoček (aminokyselinové zbytky 577-585, 615-623 a 633-641). Zaměněny byly zejména záporně nabité aminokyselinové zbytky aspartátu a glutamátu, které by se mohly podílet na koordinaci vápenatých iontů a zbytky glycinu, které jsou nezbytné pro vytvoření ostrého ohybu β-otočky. Po expresi a purifikaci mutantních variant byla stanovena biologická aktivita pomocí hemolytické eseje na beraních erytrocytech. Výsledky prokázaly, že substituce zbytku aspartátu za objemný zbytek tryptofanu (D620W, D638W) nebo trojitá hydrofobní substituce (N619L+D620L+D621L) vede k úplné ztrátě hemolytické aktivity. Tento jev...Kingella kingae is a significant pediatric bacterial pathogen, whose key virulence factor is the cytotoxin RtxA, which belongs to the family of RTX toxins. The transition of the inactive protoxin into its biologically active form requires a post-translational modification by the specific acyltransferase RtxC, followed by the binding of calcium ions to the RTX domain. The purpose of this bachelor's thesis was to investigate the recently predicted calcium-binding sites located directly within the acylated segment of the RtxA toxin, thereby expanding the knowledge of its folding mechanism into an active conformation in the extracellular environment. Using site-directed mutagenesis, 15 mutant variants of the RtxA toxin were prepared with point mutations in the regions of predicted β-turns (amino acid residues 577-585, 615- 623 and 633-641). The substitutions primarily targeted negatively charged aspartate and glutamate residues, which could participate in the coordination of calcium ions, as well as glycine residues, which are essential for the formation of the sharp bend of the β-turn.
