[eng][cze] With next-generation sequencing becoming more commonly used, new causes for OXPHOS system defects in patients rapidly emerge. All these defects are worth studying whether for greater understanding of the molecular machinery or in attempt to find an effective treatment for the patients suffering from them. However, gathering samples from each and every patient is often not feasible for numerous reasons. In this study we generate overexpression based HEK293 models, characterize them, and compare them to wild-type HEK293 cells. Finally, we look for similarities between data from the mutant model cell lines and data published from patient samples.S lepší dostupností nové generace sekvenačních technik se výrazně zrychluje nalézání mutací v DNA vedoucích k defektům v OXPHOS systému. Všechny tyto defekty je třeba studovat, ať už kvůli lepšímu pochopení molekulárních interakcí, nebo snaze nalézt léčbu pro pacienty trpící těmito defekty. Bohužel, získat vzorky ze všech těchto pacientů často není z mnoha odlišných důvodů možné. Proto jsme se v této práci rozhodli vytvořit modelové HEK293 buněčné linie na základě nadměrné exprese genu, zkoumat je a porovnat s divokou linií. Nakonec jsme srovnali data z experimentů na HEK293 modelech s již publikovanými daty z pacientů.
