Rovnováha mezi příjmem a výdejem energie je klíčová pro regulaci energetické homeostázy a pro metabolické zdraví organismu. Jednou z cest, jak tělo energii vydává, jsou termogenní mechanismy, jejichž lepší porozumění by mohlo pomoci v cestě v boji proti obezitě a metabolickým dysbalancím. Tato práce prohlubuje poznání v problematice termogenních mechanismů jiných než je termogeneze pomocí proteinu UCP1, jehož funkce je u dospělých lidí oslabena. Cílem práce bylo poznat vliv rozvoje termogenních mechanismů u UCP1-KO myší na klidový metabolismus, dále pak na energetický výdej spojený s fyzickou aktivitou, na svalový stah a na tělesnou teplotu při běhu. Část práce je věnována také optimalizaci podmínek a protokolů pro tato měření. Výsledky ukazují vliv rozvoje termogenních mechanismů na klidový metabolismus myši. Klíčovým výstupem je především pozorovaný vliv rozvoje alternativních mechanismů netřesové termogeneze na nárůst energetického výdeje myši při běhu, na sílu svalového stahu a na tělesnou teplotu během fyzické aktivity. Klíčová slova: svalová netřesová termogeneze, metabolismus, nepřímá kalorimetrie, cvičení, termoregulace The balance between energy intake and expenditure is crucial for regulating energy homeostasis and for the body's metabolic health. One of the ways the body expends energy is through thermogenic mechanisms; a better understanding of these mechanisms could help in the fight against obesity and metabolic dysbalances. This study deepens our understanding of thermogenic mechanisms other than those mediated by the UCP1 protein, whose function is impaired in adult humans. The aim of this study was to investigate the effect of the development of thermogenic mechanisms in UCP1-KO mice on resting metabolism, as well as on energy expenditure associated with physical activity, muscle contraction, and body temperature during running. Part of the study is also devoted to optimizing the conditions and protocols for these measurements. The results demonstrate the influence of the development of thermogenic mechanisms on resting metabolism in mice. A key finding is the observed influence of the development of alternative mechanisms of non-shivering thermogenesis on the increase in energy expenditure during running, on muscle contraction force, and on body temperature during physical activity.
