Josef Krátký, Ph.D. Název diplomové práce: Studium účinků nových potenciálních anthelmintik Haemonchus contortus je krev sající hlístice, způsobující onemocnění zvané haemonchóza. Poškozuje zdraví a snižuje užitkovost hospodářských zvířat, hlavně malých přežvýkavců. Haemonchóza je primárně léčena anthelmintiky. Se stále se rozšiřující rezistencí hlístic k anthelmintikům, ale léčiva přestávají být účinná. Z toho důvodu je nutné hledat nová léčiva, která mají jiný mechanismus působení a která by byla účinná i na rezistentní kmeny. Cílem této diplomové práce bylo testování potenciální anthelmintické účinnosti nových derivátů kyseliny benzhydroxamové (OMK421, OMK422, OMK423 a OMK424) na larvách a dospělcích H. contortus kmene IRE, který je rezistentní na benzimidazolová anthelmintika. Za tímto účelem byla z trusu infikovaných ovcí získána vajíčka a následně kultivovány larvy. Ze slezu byli vyizolováni dospělci H. contortus a roztříděni podle pohlaví. Anthelmintická účinnost nových derivátů kyseliny benzhydroxamové byla zjišťována sledováním vlivu těchto látek na viabilitu odpláštěných larev xL3 a na viabilitu samců a samic pomocí... Josef Krátký, Ph.D. Title of Diploma Thesis: A Study of the effects of the new potential anthelmintics Haemonchus contortus is a blood-sucking nematode that causes a disease called haemonchosis. It compromises the health and reduces the productivity of livestock, particularly small ruminants. Haemonchosis is primarily treated with anthelmintics. However, with the ever-increasing resistance of nematodes to anthelmintics, these drugs are becoming less effective. For this reason, it is necessary to search for new drugs that have a different mechanism of action and would be effective even against resistant strains. The aim of this thesis was to test the potential anthelmintic efficacy of new benzhydroxamic acid derivatives (OMK421, OMK422, OMK423, and OMK424) against larvae and adults of H. contortus strain IRE, which is resistant to benzimidazole anthelmintics. For this purpose, eggs were collected from the feces of infected sheep and the larvae were subsequently cultured. Adult H. contortus were isolated from the fleas and sorted by sex.
