Patologické hromadění tau proteinu a α-synukleinu představuje jeden ze zásadních mechanismů neurodegenerativních onemocnění, přičemž stále více poznatků poukazuje na to, že tyto proteiny mohou v nervové tkáni podléhat prionům podobnému šíření. V centru pozornosti tak stojí nejen mechanismy chybného sbalení těchto proteinů, jejich agregace a vzniku toxických oligomerů, ale také mechanismy mezibuněčného přenosu, topografie šíření patologie a heterogenita klinického obrazu. Významnou roli v tomto kontextu hraje i cross-seeding tau proteinu a α-synukleinu, který může přispívat ke vzniku smíšených proteinopatií a k variabilitě průběhu onemocnění. Experimentální modely i humánní data podporují představu, že tento koncept může sloužit jako užitečný rámec pro pochopení progrese neurodegenerativních onemocnění. Zároveň však ukazují jeho důležité limity a potvrzují, že tau a α-synuklein nelze ztotožňovat s klasickými infekčními priony. The pathological accumulation of tau protein and α-synuclein represents one of the key mechanisms underlying neurodegenerative diseases, and a growing body of evidence suggests that these proteins may undergo prion-like propagation in neural tissue. The focus is thus not only on the mechanisms of misfolding of these proteins, their aggregation, and the formation of toxic oligomers, but also on the mechanisms of intercellular transmission, the topography of disease spread, and the heterogeneity of the clinical picture. Cross-seeding of tau protein and α-synuclein also plays a significant role in this context, as it may contribute to the development of mixed proteinopathies and to the variability in disease progression. Both experimental models and human data support the idea that this concept provides a useful framework for understanding the progression of neurodegenerative diseases. At the same time, however, they highlight its significant limitations and confirm that tau and α-synuclein cannot be equated with classical infectious prions.
