[cze][eng] Práce hodnotí vliv chladové aklimace a krevních sér na toleranci kardiomyocytů vůči ischemicko-reperfuznímu (I-R) poškození. Chladová aklimace prokazatelně snižuje velikost infarktu v jeho akutní i chronické formě, přičemž tato protekce je doložena v obou režimech i u izolovaných kardiomyocytů. Chladová aklimace je též známa zvýšenou formou bioaktivních látek, které se uvolňují do krve. Vliv těchto bioaktivních látek na izolované kardiomyocyty u kontrolních a chladově aklimovaných potkanů dosud nebyl studován. Cílem této práce bylo zjistit vliv sekretomu od kontrolních i chladově aklimovaných potkanů na viabilitu izolovaných kardiomyocytů. Experimentální část sledovala účinky různých typů sér a závislost kardioprotektivního efektu na dostupnosti energetického substrátu. Izolované potkaní kardiomyocyty byly in vitro vystaveny hypoxii a reoxygenaci za účelem simulace I-R poškození. Výsledky práce ukazují vliv chladové adaptace organismu a specifických parakrinních faktorů na viabilitu buněk po ischemicko-reperfuzním poškození. Klíčová slova: Kardioprotekce, chladová aklimace, izolované kardiomyocyty, sekretom, potkanThis thesis evaluates the effect of cold acclimation and blood sera on the tolerance of cardiomyocytes to ischemia-reperfusion (I-R) injury. Cold acclimation demonstrably reduces infarct size in both its acute and chronic forms, and this protection has been documented in both regimens in isolated cardiomyocytes as well. Cold acclimation is also known for elevated levels of bioactive substances released into the bloodstream. The effect of these bioactive substances on isolated cardiomyocytes from control and cold-acclimated rats has not yet been studied. The aim of this study was to determine the effect of the secretome from control and cold-acclimated rats on the viability of isolated cardiomyocytes. The experimental part investigated the effects of various types of sera and the dependence of the cardioprotective effect on energy substrate availability. Isolated rat cardiomyocytes were subjected to in vitro hypoxia and reoxygenation to simulate I-R injury. The results of this work demonstrate the impact of the organism's cold adaptation and specific paracrine factors on cell viability following ischemia-reperfusion injury.
