[eng][cze] Cellular growth is mainly controlled by the mTORC1 signaling pathway, which acts as a master regulator by sensing the availability of essential amino acids. These amino acids are detected by a specific set of cytosolic sensors (Sestrin2, CASTOR1, and SAMTOR), which interact with GATOR protein complexes that integrate the signal and, in the presence of sufficient amino acids, induce cellular growth by activating mTORC1 kinase. This thesis aims to describe how these individual amino acid sensors recognize various ligands, such as leucine and arginine, and to provide a precise structural description of their interactions with GATOR2 and GATOR1 complexes. Finally, this thesis discusses the consequences of pathway dysregulation for human health, outlining its critical role in various metabolic disorders.Buněčný růst je primárně řízen signální dráhou mTORC1, která působí jako hlavní regulátor tím, že detekuje dostupnost esenciálních aminokyselin. Tyto aminokyseliny jsou rozpoznávány specifickou sadou cytosolových senzorů (Sestrin2, CASTOR1 a SAMTOR), které interagují s proteinovými komplexy GATOR. Tyto komplexy integrují signál a za přítomnosti dostatečného množství aminokyselin indukují buněčný růst aktivací kinázy mTORC1. Tato bakalářská práce se zaměřuje na popis toho, jak jednotlivé senzory aminokyselin rozpoznávají své ligandy, jako leucin a arginin, a poskytuje přesný strukturní popis interakcí s komplexy GATOR2 a GATOR1. Na závěr tato práce pojednává o následcích dysregulace této dráhy pro lidské zdraví a nastiňuje její kritickou roli u různých metabolických poruch.
