[cze][eng] Radioterapie je klíčovou metodou v léčbě onkologických onemocnění, její účinnost je však často limitována poškozením zdravých tkání. Vývoj nových, bezpečných radioprotektivních látek je proto zásadní pro zlepšení terapeutického indexu. V rámci této práce byla syntetizována série nových sloučenin vycházejících z 1-(2-hydroxyethyl)piperazinového skeletu, který byl na základě předchozích studií vybrán pro svůj potenciální radioprotektivní účinek. Cílem práce byla příprava deseti nových derivátů pomocí dvoukrokové syntézy, která zahrnovala reakci epibromhydrinu se substituovaným anilinem a následnou nukleofilní substituci vzniklého meziproduktu 1-(2- hydroxyethyl)piperazinem. Celkově bylo úspěšně připraveno a charakterizováno sedm finálních produktů s celkovými výtěžky v rozmezí 8-68 %. Průběh syntézy byl silně ovlivněn sterickými a elektronovými vlastnostmi substituentů na anilinovém jádře, přičemž substituce v ortho poloze se ukázala jako problematická a vedla k rozkladu meziproduktů. U všech připravených sloučenin byla provedena in silico analýza farmakokinetických vlastností. Výsledky ukázaly, že všechny nové sloučeniny mají příznivý teoretický profil pro perorální podání, protože splňují Lipinského pravidlo pěti, vykazují vysokou předpokládanou absorpci v gastrointestinálním traktu a neprocházejí...Radiotherapy is a key method in the treatment of oncological diseases; however, its effectiveness is often limited by damage to healthy tissues. The development of new, safe radioprotective agents is therefore essential for improving the therapeutic ratio. In this work, a series of new compounds based on the 1-(2-hydroxyethyl)piperazine scaffold was synthesized, which was selected for its potential radioprotective effect based on previous studies. The aim of the work was the preparation of ten new derivatives using a two-step synthesis, which included the reaction of epibromohydrin with substituted aniline followed by nucleophilic substitution of the resulting intermediate with 1-(2-hydroxyethyl)piperazine. Overall, seven final products were successfully prepared and characterized, with overall yields ranging from 8 to 68%. The course of the synthesis was strongly influenced by the steric and electronic properties of the substituents on the aniline core, while substitution in the ortho position proved to be problematic and led to the decomposition of the intermediates. An in silico analysis of pharmacokinetic properties was performed for all prepared compounds.
